不同方法提取阴道加德纳菌细菌基因组DNA的比较

目的对阴道加德纳菌基因组DNA几种不同的提取方法进行比较,以利于细菌性阴道病的研究。方法采用几种不同方法提取阴道加德纳菌基因组DNA,对不同方法提取的细菌基因组DNA进行定量检测,以评价不同方法的提取效率。结果加德纳菌基因组DNA提取以改良法提取率较高,酚-氯仿抽提法、醋酸钾法次之,碱裂解煮沸法和溶菌酶煮沸法提取效率较差,加热煮沸法提取效率低。结论改良法提取GV基因组DNA效率较好,适用于临床应用。

石蜡包埋淋巴组织三种微量DNA提取方法的比较

目的为寻找一种高效、简便、实用的石蜡组织中DNA提取方法,本研究比较了三种不同的DNA提取方法对DNA质量的影响。方法选取淋巴相关组织的蜡块21例。三种DNA提取方法分别是简单消化煮沸法,酚/氯仿提纯法和试剂盒法。通过提取DNA的浓度和纯度、PCR扩增保守序列β-globin等方法,比较三种DNA提取方法的优劣。结果试剂盒法提取样品OD260/OD280平均比值位于1.61.8之间;提纯法位于1.31.5之间;简单消化煮沸法位于1.21.3之间。三种方法中,试剂盒法提取的DNA最为稳定 ...

为了建立一种检测水中铜绿微囊藻浓度的定量PCR方法,分别用SDS裂解法和DNeasy Blood & Tissue Kit试剂盒提取标准品的DNA并建立标准曲线.结果表明采用DNeasy Blood & Tissue Kit试剂盒提取的标准品DNA具有很好的重复性,经定量PCR扩增得到的标准曲线R2为0.968,可以用来检测样品中铜绿微囊藻的浓度,为建立一套长期有效的测定水中铜绿微囊藻浓度的监测体系提供了技术参考.

三种简易提取全血基因组DNA方法的比较

目的比较三种快速、简便、适用、低成本的从全血中提取基因组DNA的方法。方法选用快速氯仿-异戊醇法、简化盐析法、改良碘化钾法直接从全血中提取基因组DNA。结果改良碘化钾法提取的DNA纯度及含量最高,快速氯仿-异戊醇法次之,简化盐析法则相对较低。结论改良碘化钾法为较好的提取基因组DNA的方法。

石蜡包埋组织中DNA提取方法的比较

目的通过比较两种从石蜡包埋组织中提取基因组DNA的方法,确定一种高效、经济而简便的方法指导实际工作。方法按照两种方法(脱蜡法与非脱蜡法)提取DNA,比较所提DNA的质量,PCR扩增β-Globin基因作为基因分析的对照扩增,并对其中非脱蜡法进行改良。结果两种方法所提取的DNA均能达到相关分子学实验的要求,PCR扩增带清晰,结果较为相似。结论非脱蜡法也能提取出适用于PCR技术要求的DNA,方法简便、经济。
