Cytosine-5-methyltransferases add aldehydes to DNA

Nature Chemical Biology, 2009
Targeted methylation of cytosine residues by S-adenosylmethionine-dependent DNA methyltransferases modulates gene expression in vertebrates. Here we show that cytosine-5-methyltransferases catalyze reversible covalent addition of exogenous aliphatic aldehydes to their target residues in DNA, thus yielding corresponding 5-hydroxyalkylcytosines.

Mammalian DNA (cytosine-5) methyltransferases and their expression

Clinical Immunology, 2003
Two classes of functional DNA (cytosine-5) methyltransferases have been discovered in mammals to date. One class methylates the unmodified DNA and is designated as the de novo enzyme, whereas the other maintains the methylation status of the daughter strand during DNA replication and thus is referred to as a maintenance DNA methyltransferase.

Recognition of foldback DNA by the human DNA (cytosine-5-) methyltransferase

Biochemistry, 1992
In order to specify the recognition requirements of the human DNA (cytosine-5-)-methyltransferase, two isomeric 48mers were synthesized so as to link a long block of DNA with a shorter complementary block of DNA through a tether consisting of five thymidine residues.

Recognition of unusual DNA structures by human DNA(cytosine-5)methyltransferase

Journal of Molecular Biology, 1991
The symmetry of the responses of the human DNA (cytosine-5)methyltransferase to alternative placements of 5-methylcytosine in model oligodeoxynucleotide duplexes containing unusual structures has been examined. The results of these experiments more clearly define the DNA recognition specificity of the enzyme. A simple three-nucleotide recognition motif

Human DNA (cytosine-5)-methyltransferases: a functional and structural perspective for epigenetic cancer therapy

Biochimie, 2017
Epigenetic modifications modulate chromatin states to regulate gene expression. Among them, DNA methylation and histone modifications play a crucial role in the establishment of the epigenome. In cancer, these epigenetic events may act in concert to repress tumor suppressor genes or promote oncogenic pathways.

Molecular Cloning, Expression, and Characterization of Starfish DNA (Cytosine-5)-methyltransferases

Bioscience, Biotechnology, and Biochemistry, 2012
To determine whether and if so how a DNA methylation-dependent epigenetic mechanism for transcriptional gene silencing functions in Echinoderms, we cloned and sequenced dnmt1 and dnmt3 cDNAs of the starfish Asterina pectinifera. Since the Strongylocentrotus purpuratus genome has only two loci of DNA (cytosine-5)-methyltransferase genes encoding Dnmt1 ...

Isolation and characterisation of DNA cytosine 5-methyltransferase from human placenta

Biochimica et Biophysica Acta (BBA) - Gene Structure and Expression, 1983
DNA cytosine 5-methyltransferase has been extensively purified (about 2600-fold) from the soft tissue of human placenta by chromatography on DEAE-cellulose and hydroxyapatite, and by an affinity step on agarose-immobilized S-adenosylhomocysteine. The isolated enzyme has a molecular weight of 135,000 and methylates DNA from various sources in native and

DNA (cytosine-5)-methyltransferase 3B - [Isoform 1]

Targeted Protein Database, 2008
A DNMT3B (DNMT3B , DNA (cytos ine-5)methyltransferase 3B; Swiss-Prot accession number: Q9UBC3) gene deletion construct in which exons 16–19 were deleted was used to generate Dnmt3b-null ES cells and animals. The deleted region encodes the highly conserved PC (proline cysteine) and ENV (glutamic acid, asparagine, valine) motifs that are crucial for ...

Mammalian (cytosine-5) methyltransferases cause genomic DNA methylation and lethality in Drosophila

Nature Genetics, 1999
CpG methylation is essential for mouse development as well as gene regulation and genome stability. Many features of mammalian DNA methylation are consistent with the action of a de novo methyltransferase that establishes methylation patterns during early development and the post-replicative maintenance of these patterns by a maintenance ...

DNA (Cytosine-5) Methyltransferase Inhibitors: A Potential Therapeutic Agent for Schizophrenia

Molecular Pharmacology, 2007
In this issue of Molecular Pharmacology, Kundakovic et al. (p. 644) present compelling evidence suggesting that the promoters for reelin and GAD67 are coordinately regulated. The regulation occurs at the level of DNA (cytosine-5) methylation. Moreover, the authors present evidence suggesting that pharmacologic inhibition of DNA methyltransferase ...
