No evidence for AID/MBD4-coupled DNA demethylation in zebrafish embryos. [PDF]

open access: yesPLoS ONE, 2014
The mechanisms responsible for active DNA demethylation remain elusive in Metazoa. A previous study that utilized zebrafish embryos provided a potent mechanism for active demethylation in which three proteins, AID, MBD4, and GADD45 are involved.

Mechanical Activation of Piezo1 Drives Osteoarthritis Through Kdm5c‐Mediated Epigenetic Silencing

open access: yesAdvanced Science, EarlyView.
Excessive mechanical stress activates Piezo1, triggering Ca2+‐dependent cytoskeletal forces that deform the nucleus and reduce H3K4me3. Kdm5c demethylates H3K4me3 at Col2a1 and Runx3 promoters. Kdm5c knockout rescues degradation. Repurposed telmisartan directly inhibits Kdm5c, blocking this axis and showing disease‐modifying efficacy in mouse OA models

NF-kappaB mediates Gadd45beta expression and DNA demethylation in the hippocampus during fear memory formation.

open access: yesFrontiers in Molecular Neuroscience, 2015
Gadd45-mediated DNA demethylation mechanisms have been implicated in the process of memory formation. However, the transcriptional mechanisms involved in the regulation of Gadd45 gene expression during memory formation remain unexplored.

Reprogramming of fibroblast cells to totipotent state by DNA demethylation

open access: yesScientific Reports, 2023
Many attempts have been made to induce high-quality embryonic stem cells such as pluripotent stem cells and totipotent stem cells, but challenges remain to be overcome such as appropriate methods and sources.

RBM12 Maintains Glioma Stem Cells by Activating Amino Acid‐Dependent mTORC1 Signaling via SLC7A5 mRNA Stabilization

open access: yesAdvanced Science, EarlyView.
This study shows that in glioma stem cells (GSCs), RBM12 recruits ALKBH5 to remove m6A from SLC7A5 transcripts, thereby enhancing mRNA stability, which elevates large neutral amino acid (LNAA) levels and activates mTORC1, promoting GSC proliferation, self‐renewal, and tumor growth.

DNA demethylation is involved in nitric oxide-induced flowering in tomato

open access: yesJournal of Integrative Agriculture
Flowering is one of the most important phenological periods, as it determines the timing of fruit maturation and seed dispersal. To date, both nitric oxide (NO) and DNA demethylation have been reported to regulate flowering in plants.

Oxidative Demethylation of DNA

open access: yesThe FASEB Journal, 2007
Oxidative demethylation of DNA Non‐enzymatic methylation of DNA by alkylating agents such as MMS occurs at several sites, but not at the 5 position of cytosine. Instead, the main cytotoxic lesions are 3‐methyladenine, and in single‐stranded regions of DNA also 1‐methyladenine and 3‐methylcytosine.

Liposomal Delivery of L‐2‐Hydroxyglutarate for Targeting Epigenetic Dysregulation in Osteoarthritis

open access: yesAdvanced NanoBiomed Research, EarlyView.
Osteoarthritis involves cartilage degeneration, inflammation, and epigenetic dysregulation. A liposomal formulation of the TET1 inhibitor L‐2‐hydroxyglutarate to overcome rapid joint clearance was developed. The formulation showed good physicochemical properties, reduced inflammatory and catabolic gene expression in chondrocytes, and alleviated ...

Aberrant DNA methylation of Tgfb1 in diabetic kidney mesangial cells

open access: yesScientific Reports, 2018
Epigenetic modulation may underlie the progression of diabetic nephropathy (DN). Involvement of TGFB1 in mesangial fibrosis of DN led us to hypothesize that Tgfb1 DNA demethylation contributes to progression of DN.

TET-mediated DNA hydroxymethylation is negatively influenced by the PARP-dependent PARylation

open access: yesEpigenetics & Chromatin, 2022
Background Poly(ADP-ribosyl)ation (PARylation), a posttranslational modification introduced by PARP-1 and PARP-2, has first been implicated in DNA demethylation due to its role in base excision repair. Recent evidence indicates a direct influence of PARP-
