Volunteer infection studies using the induced blood stage malaria (IBSM) model have been shown to facilitate antimalarial drug development. Such studies have traditionally been undertaken in single‐dose cohorts, as many as necessary to obtain the dose ...

Degradation mechanism of the von Willebrand factor A2 domain by nattokinase
Nattokinase, a natto‐derived protease, exhibits potent antithrombotic effects. This study demonstrates that nattokinase directly cleaves the von Willebrand factor (vWF) A2 domain in vitro. Unlike the native regulator ADAMTS13, nattokinase degrades folded vWF independently of shear stress.

Deep sedation with midazolam and propofol in children undergoing ambulatory magnetic resonance imaging of the brain [PDF]
Backround/Aim. Sedation is necessary in children undergoing magnetic resonance imaging (MRI) to ensure motionless. The success of sedation is typically measured by two factors: safety (lack of adverse events) and effectiveness of the procedure ...

Proteostasis and the gut microbiota play a key role in shaping host physiology. Microbiota‐derived metabolites, vitamins, and RNA modulate host proteostasis. Findings from model systems, including C. elegans, indicate microbes can either stabilize or disrupt host proteostasis.

Relationship Between Response and Dose in Published, Contemporary Phase I Oncology Trials. [PDF]
BackgroundAs progress continues in oncology drug development, this study aimed to examine whether the previously established association between drug dose and efficacy in the era of cytotoxic therapies remains true in today's phase I dose-escalation ...

Design and analysis strategies for robust microbiome ageing research
The gut microbiome changes with age and associates with age‐related morbidity and mortality, establishing it as a potential biomarker and intervention target for ageing. Realising this potential requires methodological rigour, yet distinguishing biological signals from methodological artefacts remains challenging across cohorts. This review provides an

Relationship between drug dose and drug effect
It is generally assumed that there exists a well-defined relationship between drug dose and drug effect and that this can be expressed by a dose-response curve.

Evidence for a curvilinear dose-response relationship between avoidance coping and drug use problems among women who experience intimate partner violence [PDF]
Women who experience intimate partner violence (IPV) are at heightened risk for drug use problems. While prevailing models of drug use suggest that IPV-exposed women use drugs in an effort to escape or avoid negative affect, a dearth of literature has examined the role of avoidance coping in drug use problems within this population.

Investigating transcription factor dynamics in health and disease using FRAP
FRAP analysis of GFP‐tagged transcription factors reveals how molecular mobility and target engagement change in response to drug treatment. By combining live‐cell imaging, quantitative model fitting, and statistical analysis, this approach uncovers transcription factor dynamics linked to disease mechanisms, providing a powerful framework for ...
