Controllable Generation of Pathogen‐Specific Antimicrobial Peptides Through Knowledge‐Aware Prompt Diffusion Model

open access: yesAdvanced Science, EarlyView.
KPPepGen is a knowledge‐aware prompt diffusion model for the controllable generation of pathogen‐specific AMPs. It outperforms existing methods and is capable of simultaneously generating peptides for distinct pathogens, demonstrating superior performance with favorable properties and docking efficacy.

Engineered Fusion Enzyme‐Mediated Non‐Consecutive Cyclization‐Glycosylation Enables Heterologous Synthesis of Antifungal Enfumafungin

open access: yesAdvanced Science, EarlyView.
The artificial biosynthetic pathway potentially involved in biosynthesis of enfumafungin is first reconstructed in an optimized Aspergillus oryzae chassis with the increased site‐specific integration efficiency and shortened time for marker recycling, which requires an artificial fusion enzyme EfuA(TC)FsoA(GT).

Biofilm formation and drug susceptibility of biofilm Candida spp. clinically isolated from nasopharyngeal cancer patients in Vietnam

open access: yesIranian Journal of Microbiology
Background and Objectives: The biofilm formation has been widely recognized as one of the main mechanisms of antimicrobial resistance development in microorganisms. However, few studies are focusing on this phenomenon in Candida spp.

Alternative Models for Anticancer Drug Discovery From Natural Products Using Binary Tumor‐Microenvironment‐on‐a‐Chip

open access: yesAdvanced Science, EarlyView.
This study presents a binary tumor‐microenvironment‐on‐a‐chip (T‐MOC) system incorporating multicellular tumor spheroids (MCTs) as an alternative preclinical platform to evaluate the efficacy of anticancer natural products. The T‐MOC model reproduces in vivo drug delivery barriers and physiological conditions, enabling morphological analysis to predict

Repurposing Cardiac Glycosides to Potentiate CD47 Blockade through Calreticulin‐mediated Phagocytic Effects for Lung Cancer Treatment

open access: yesAdvanced Science, EarlyView.
Repurposing cardiac glycosides (CGs) significantly strengthens the efficacy of anti‐CD47 antibody for lung cancer treatment by boosting macrophage phagocytosis. This novel combination blocks the “don't eat me” signal and increases dual “eat‐me” signals, Fc‐FcγR and cell surface expression of calreticulin (ecto‐CRT).

Update on the Fungal Biofilm Drug Resistance and Its Alternative Treatment

open access: yesJournal of Biosciences and Medicines, 2016
Candida albicans (C. albicans) and Aspergillus fumigatus (A. fumigatus) are the two main pathogens in the clinical setting to cause serious, sometimes, lethal fungal infections. Immunocompromised patients fall victims to these fungi, with a mortality rate rising drastically over the past decades.

Transcriptional control of drug resistance, virulence and immune system evasion in pathogenic fungi: a cross-species comparison.

open access: yesFrontiers in Cellular and Infection Microbiology, 2016
Transcription factors are key players in the control of the activation or repression of gene expression programs in response to environmental stimuli. The study of regulatory networks taking place in fungal pathogens is a promising research topic that ...

Hydrogen‐Deuterium Exchange Mass Spectrometry for Molecular Glue Characterization

open access: yesAdvanced Science, EarlyView.
Molecular glues that stabilize protein–protein interactions are challenging to identify. Here, hydrogen‐deuterium exchange mass spectrometry can show and reveal gluing mechanisms that vary between protein binding partners. Abstract Molecular glues are powerful bioactive molecules that stabilize protein–protein interactions.

Identification of a Pair of Linear or Cyclic Naturally Inspired Bifunctional Lipopeptide Antibiotics That Overcome Antimicrobial Resistance

open access: yesAdvanced Science, EarlyView.
The underexplored bacterial genus Aquimarina harbors a large biosynthetic potential for discovering new antibiotics. Here, the authors identify a pair of linear or cyclic bifunctional cationic lipopeptide antibiotics, aquicidine L and C4, inspired by the aquicidine gene cluster from Aquimarina.
