Protective effect of aqueous extract of Pleurotus eous against Dalton's lymphoma ascites tumor in mice

open access: yesJournal of Agriculture and Food Research, 2023
The antitumor efficacy of Pleurotus eous aqueous extract (PEAE) was investigated both in vitro and in vivo. The MTT test was used to determine in vitro cytotoxicity against the Dalton's Lymphoma Ascites (DLA) cell line.

Protein-Ligand Scoring with Convolutional Neural Networks [PDF]

open access: yes, 2016
Computational approaches to drug discovery can reduce the time and cost associated with experimental assays and enable the screening of novel chemotypes. Structure-based drug design methods rely on scoring functions to rank and predict binding affinities and poses.

Progesterone Receptor Chaperone Complex–Based High-Throughput Screening Assay

open access: yesJournal of biomolecular screening, 2015
Hsp90 and its co-chaperones are known to be important for cancer cell survival. The N-terminal inhibitors of Hsp90 that are in ongoing clinical trials as antitumor agents have unfortunately shown disappointing efficacies in the clinic.

Phase 1 dose escalation study of the allosteric AKT inhibitor BAY 1125976 in advanced solid cancer-Lack of association between activating AKT mutation and AKT inhibition-derived efficacy [PDF]

open access: yes, 2019
This open-label, phase I first-in-human study (NCT01915576) of BAY 1125976, a highly specific and potent allosteric inhibitor of AKT1/2, aimed to evaluate the safety, pharmacokinetics, and maximum tolerated dose of BAY 1125976 in patients with advanced ...

TRPM8 levels determine tumor vulnerability to channel agonists

open access: yesMolecular Oncology, EarlyView.
TRPM8 is a Ca2+ permissive channel. Regardless of the amount of its transcript, high levels of TRPM8 protein mark different tumors, including prostate, breast, colorectal, and lung carcinomas. Targeting TRPM8 with channel agonists stimulates inward calcium currents followed by emptying of cytosolic Ca2+ stores in cancer cells.

Activation of MAPK signalling results in resistance to saracatinib (AZD0530) in ovarian cancer [PDF]

open access: yes, 2017
SRC tyrosine kinase is frequently overexpressed and activated in late-stage, poor prognosis ovarian tumours, and preclinical studies have supported the use of targeted SRC inhibitors in the treatment of this disease.

The anticancer effect of the HDAC inhibitor belinostat is enhanced by inhibitors of Bcl‐xL or Mcl‐1 in ovarian cancer

open access: yesMolecular Oncology, EarlyView.
The pan‐HDAC inhibitor belinostat increases the expression of the pro‐apoptotic proteins Bim, Puma, and Noxa and induces apoptosis in ovarian cancer cell lines and patient‐derived tumor organoids when used at high concentrations. Moreover, inhibiting the anti‐apoptotic proteins Bcl‐xL or Mcl‐1 sensitizes these preclinical models to the cytotoxic effect

On the discovery of a potential survivin inhibitor combining computational tools and cytotoxicity studies

open access: yesHeliyon, 2019
Survivin protein is a metalloprotein member of the inhibitors of apoptosis proteins family, involved in the regulation of programmed cell death. Due to the recent development of antitumor therapies having survivin as molecular target, several strategies ...

Moving microfluidics ahead: Extending capabilities, accessibility, and applications [PDF]

open access: yesarXiv, 2018
Paul Blainey is professor of Biological Engineering at MIT. In this contribution he describes three microfluidic technologies that he and his team has developed to extend the capability, accessibility, and applications of microfluidics: (1) Integrated microfluidic sample preparation for genomic assays, (2) hydrogel-based microfluidics for single-cell ...

Suillus collinitus methanolic extract increases p53 expression and causes cell cycle arrest and apoptosis in a breast cancer cell line [PDF]

open access: yes, 2012
In the present work, methanolic, ethanolic and boiled water extracts of Suillus collinitus were chemically characterized and further submitted to evaluation of their bioactive properties (antioxidant potential and cytotoxic activity in tumor cell lines).
