Multi‐Targeting Carnosic Acid Kills Drug‐Resistant Helicobacter pylori With Narrow‐Spectrum Activity

open access: yesAdvanced Science, EarlyView.
Carnosic acid selectively eradicates drug‐resistant Helicobacter pylori through a multi‐targeted mechanism involving urease inhibition, membrane disruption, and biofilm eradication. It exhibits no detectable resistance, potent in vivo efficacy, and minimal impact on gut microbiota, positioning it as a promising narrow‐spectrum lead compound against ...

Tandem Duplication‐Driven Neofunctionalization of UDP‐Glycosyltransferases Shapes the Diversification of Triterpenoid Saponins in the Cucurbitaceae

open access: yesAdvanced Science, EarlyView.
We present a chromosome‐level genome assembly of Siraitia grosvenorii and, through comparative genomics, uncover a conserved UGT73 tandem array driving triterpenoid saponin diversification in Cucurbitaceae. Crystalized SgUGT73AM30 further reveals the regioselectivity mechanism underlying its catalytic activity.

Generic competition and price developments in the USA, Germany and Switzerland (2007–2023): a longitudinal observational study

open access: yes
Introduction Drug expenditure has increased over the past years in the USA and Europe, mainly driven by originator drugs. Generic drugs have a substantial cost-saving potential. We analysed the number of generic competitors entering the market over time,

Beyond Potency: Emerging Determinants and Optimization Strategies Enhancing Therapeutic Efficacy of Adult Stem Cells

open access: yesAdvanced Science, EarlyView.
Adult stem cell therapy requires more than high in vitro potency. This review proposes a systems framework in which cell‐intrinsic programs, instructive microenvironmental cues, and pre‐/post‐delivery engineering are co‐designed under standardized translational rules.

Nano‐Backpack Engineered Probiotics with Reactive Oxygen Species–Responsive Tungsten Release for Synergistic Therapy of Inflammatory Bowel Disease

open access: yesAdvanced Science, EarlyView.
An integrated probiotic system is engineered to execute a programmed therapeutic cascade against inflammatory bowel disease. Upon pathological signals, the system sequentially performs ROS scavenging, tungsten release, and selective pathogen suppression, thereby breaking the oxidative stress–dysbiosis cycle to remodel gut microbiota and reinforce ...

Bioinspired Passive Flow Routing to Mitigate Thrombosis in Prosthetic Heart Valves and Cardiovascular Devices

open access: yesAdvanced Science, EarlyView.
Bioinspired passive flow routing mitigates thrombosis in cardiovascular devices by redirecting a small fraction of forward flow into stagnation‐prone regions. Inspired by avian alula and aircraft slat mechanisms, strategically placed routing channels restore washout around mechanical heart valves, reducing low‐shear exposure by four orders of magnitude

Is the Rate of Generic Drug Prescribing a Fair Measure for Pay for Performance?

open access: yes
Prescription drugs are a significant and rising component of healthcare costs. To limit this expenditure and maximize cost savings, half of the pay-for-performance health programs in the US reward generic drug prescribing once the patents on brand-name ...
