Characterisation of the N‐Methyltransferase SgPsmC: Application in the Kinetic Resolution of Pyrroloindolines

open access: yesAngewandte Chemie International Edition, EarlyView.
The N‐methyltransferase SgPsmC was characterised and applied in the kinetic resolution (KR) of easily obtainable pyrroloindolines. Substitution at 3a‐position proved to be a principal determinant for the enzyme's enantioselectivity. While moving toward laboratory scale, the reaction was coupled with a methionine adenosyl transferase for in situ ...

Mechanistic Characterisation of Bacterial Terpene Synthases from Chitinophagaceae Producing Marine‐Type Diterpenes

open access: yesAngewandte Chemie International Edition, EarlyView.
Six bacterial terpene synthases are reported, one of which produces instable chitinosphaerol, which is spontaneously transformed into several known natural products requiring structural revision. A second enzyme yields thermally labile neodolabella‐1(14),2,7‐triene, which upon heating shows sequential conversion into thermocyclenes A–C.

On the Origin of Substrate Specificity of Enzymes from the Amidohydrolase Superfamily

open access: yesAngewandte Chemie International Edition, EarlyView.
Comprehensive analyses of two enzyme classes from the amidohydrolase superfamily (AHS) revealed that catalysis proceeds either via 1,4 or 1,6 nucleophilic conjugate addition and that this property defines substrate specificity. Moreover, although all substrates and products are entirely achiral, this mechanistic difference results in an inverted ...
