A colorimetric method to quantify endo-polygalacturonase activity

We report a new colorimetric assay to quantify endo-polygalacturonase activity, which hydrolyzes polygalacturonic acid to produce smaller chains of galacturonate. Some of the reported polygalacturonase assays measure the activity by detecting the appearance of reducing ends such as the Somogyi-Nelson method.

AN ENDO-POLYGALACTURONASE IN CUCUMBER FRUIT

A homogenous endo-polygalacturonase (EC 3.2.1.15,) has been isolated from mature pickling cucumbers. This is the first report of an endo-polygalacturonase in cucumbers. The enzyme was purified by three chromatography steps on Sephadex cation exchangers. The molecular weight is 35,000 daltons.

Glycan analysis of recombinant Aspergillus niger endo-polygalacturonase A

The enzyme endo-polygalacturonase A, or PGA, is produced by the fungus, Aspergillus niger, and appears to play a critical role during invasion of plant cell walls. The enzyme has been homologously overexpressed in order to provide sufficient quantities of purified enzyme for structural and biological studies.

Chromogenic substrate for endo-polygalacturonase detection in gels

Abstract A simple, sensitive zymogram technique for the detection of endo-polygalacturonase (EC 3.2.1.15) in gel slabs after electrophoresis or isoelectrofocusing was developed. This technique employs a new chromogenic substrate prepared by coupling D -galacturonan DP 10 with Ostazin Brilliant Red S-5B dye.
