TopCas: Topology‐Gated Cas12a via DNA‐RNA Chimeric Circular crRNA for Amplification‐Free Nucleic Acid Detection and Conditional Gene Editing

open access: yesAdvanced Science, Volume 13, Issue 33, 15 June 2026.
The TopCas platform utilizes an engineered DNA‐RNA chimeric circular crRNA to establish topology‐gated control over CRISPR‐Cas12a activity, enabling target‐triggered self‐catalytic amplification (preamplification‐free). This system seamlessly integrates highly sensitive molecular diagnostics with conditionally controlled gene editing, demonstrating ...

T‐Cell Exhaustion in the Tumor Microenvironment: Subcellular Dysfunction, Pan‐Cancer Characteristics, and Therapeutic Interventions

open access: yesAdvanced Science, Volume 13, Issue 33, 15 June 2026.
This study elucidates the mechanisms of subcellular multidimensional collapse in exhausted T cells. By specifically targeting the nucleus, mitochondria, and endoplasmic reticulum, strategic interventions can effectively remodel the compromised organelle network. This integrated approach drives comprehensive T cell resuscitation, ultimately establishing

Engineering of CoA‐Acylating Butyraldehyde Dehydrogenase for Enhanced 1,3‐Butanediol Production in Escherichia coli

open access: yesBiotechnology and Bioengineering, Volume 123, Issue 6, Page 1605-1614, June 2026.
ABSTRACT Microbial production of 1,3‐butanediol (1,3‐BDO) offers a renewable route to this versatile C4 chemical. However, the low performance of CoA‐acylating butyraldehyde dehydrogenase (Bld), which contains a catalytic cysteine, limits efficient production in recombinant Escherichia coli (E. coli).

A Mechanistic Model of rAAV Production in Synthetic Cell Lines

open access: yesBiotechnology and Bioengineering, Volume 123, Issue 6, Page 1684-1694, June 2026.
ABSTRACT The recombinant adeno‐associated virus (rAAV) is a widely used vector for gene therapy. Its manufacturing faces significant challenges in producing the large quantities of vectors needed for clinical applications and reducing empty particles.

USP5‐C‐MAF Axis Regulates Autophagy‐Dependent Neuronal Ferroptosis in Spinal Cord Injury Therapeutics

open access: yesCNS Neuroscience &Therapeutics, Volume 32, Issue 6, June 2026.
SCI induces significant upregulation of USP5 in neurons. The overexpressed USP5 stabilizes c‐MAF protein expression through its deubiquitinase activity, activating autophagy and accelerating the degradation of autophagic ferritin, thereby mediating neuronal ferroptosis. This ultimately leads to impaired functional recovery in SCI mice.
