Addition of sHGP-9r to facilitate endosome escape of nanocomplexes. [PDF]

open access: yes, 2015
(A, Top) Confocal microscopy images of HeLa cells pretreated with the early endosome marker-RFP, and subsequently incubated with I4R-9r PwSN carrying FITC labeled siRNA.

Rab21 recruits EEA1 and competes with Rab5 for Rabex-5 activation

open access: yesFrontiers in Cell and Developmental Biology
Rab5 is a key regulator of early endosomal traffic and fusion. It shares its localization and guanine nucleotide exchange factor Rabex-5 with the less characterized member of the Rab5 subfamily Rab21.

Exosome Production Is Key to Neuronal Endosomal Pathway Integrity in Neurodegenerative Diseases

open access: yesFrontiers in Neuroscience, 2019
Dysfunction of the endosomal–lysosomal system is a prominent pathogenic factor in Alzheimer’s disease (AD) and other neurodevelopmental and neurodegenerative disorders. We and others have extensively characterized the neuronal endosomal pathway pathology

IFITM3 Restricts Influenza A Virus Entry by Blocking the Formation of Fusion Pores following Virus-Endosome Hemifusion

open access: yesPLoS Pathogens, 2014
Interferon-induced transmembrane proteins (IFITMs) inhibit infection of diverse enveloped viruses, including the influenza A virus (IAV) which is thought to enter from late endosomes.

Targeting DNGR‐1 with Fangchinoline Elevates Dendritic Cell Antigen Cross‐Presentation‐Mediated Antitumor Immunity in Melanoma

open access: yesAdvanced Science, EarlyView.
Fangchinoline is identified as a small‐molecule DNGR‐1 modulator that enhances dendritic‐cell cross‐presentation of tumor antigens. By engaging DNGR‐1 and activating Syk–Nox2 signaling, it promotes phagosomal ROS, antigen escape, MHC‐I presentation, and CD8+ T‐cell priming, thereby strengthening antitumor immunity and sensitizing tumors to PD‐1 ...

Loss of MYO5B expression deregulates late endosome size which hinders mitotic spindle orientation [PDF]

open access: yes, 2021
Recycling endosomes regulate plasma membrane recycling. Recently, recycling endosome-associated proteins have been implicated in the positioning and orientation of the mitotic spindle and cytokinesis.

Endobody: Genetically Encodable Nanobody‐CPP Chimeras for Degradation of Membrane and Extracellular Proteins

open access: yesAdvanced Science, EarlyView.
We introduced genetically encodable, receptor‐independent nanobody‐CPP chimeras, termed endobodies, as robust and modular membrane protein degraders. Additionally, proteasome‐targeting domain (PTD)‐tethered endobody demonstrates further enhanced degradation potency.

Hemoglobin's α‐Helix‐to‐β‐Sheet Transition Enables Targeted mRNA Delivery to the Lung

open access: yesAdvanced Science, EarlyView.
Acidic heating converts hemoglobin into β‐sheet‐rich fibrils with positively charged surfaces. These hemoglobin fibrils electrostatically complex IL‐11 scFv mRNA, selectively bind circulating platelets, and hitchhike to the lung, where they are enriched in fibroblasts.

On the fate of early endosomes

open access: yesbchm, 2009
Abstract Proteins are endocytosed by various pathways into the cell. All these pathways converge at the level of the early endosome. The fate of the early endosome and how proteins are sorted into recycling and late endosomes/multi-vesicular body is a matter of debate and intense research.

Membrane Fusion‐Mediated Cytosolic Delivery of Threose Nucleic Acids via Homotypic Nanoparticles Overcomes Drug Resistance in Triple‐Negative Breast Cancer

open access: yesAdvanced Science, EarlyView.
This study introduces a biomimetic “nanofusion” platform that integrates the biostability of threose nucleic acids (TNA) with homotypic cell‐membrane cloaking to combat drug‐resistant TNBC. By leveraging a non‐canonical membrane‐fusion pathway for direct cytosolic delivery, the platform bypasses endosomal sequestration. To achieve potent AKT2 silencing
