Vitamin K epoxide reductase activity in the metabolism of epoxides

The importance of vitamin K epoxide reductase for the metabolism of a range of structurally diverse epoxides has been investigated. Vitamin K1 epoxide is reduced by rat liver microsomes at a rate of 0.47 nmoles/g liver/min. The rate of menadione oxide reduction is not significantly higher than the non-enzymatic reduction rate.

Inhibition of liver microsomal epoxide hydrase by cyproheptadine epoxide

Cyproheptadine epoxide is a competitive inhibitor or rat liver microsomal epoxide hydrase with an apparent K1-value of 0.75 mM. Cyclobenzaprine and its epoxide stimulate in vitro the activity of this enzyme, whereas cyproheptadine, carbamazepine and carbamazepine epoxide have no effect.
