The ErbB receptor family: a therapeutic target for cancer
Trends in Molecular Medicine, 2002This article reviews the current state of efforts targeting the ErbB family of tyrosine kinase receptors in cancer therapy. In particular, preliminary results will be discussed of studies of the first generation of therapeutics to enter clinical evaluation in malignant diseases.

Many such interactions are mediated by RTKs and their soluble or surface-bound ligand growth factors. It contains four distinct segments: an N-terminal EGF binding domain which is exposed at the cell surface; a short transmembrane segment; a cytoplasmic domain with protein-tyrosine kinase activity; and a C-terminal regulatory segment.
