Structural and functional analysis of Bacillus sarcosine oxidase and its activity toward cyclic imino acids

open access: yesFEBS Open Bio, EarlyView.
Sarcosine oxidase from Bacillus sp. (SoxB) recognizes l‐proline (l‐Pro), d‐proline (d‐Pro), and l‐thioproline (l‐Tpr) as minor substrates. In this study, we measured their absorption spectra, determined the crystal structures of their enzyme complexes, and performed site‐directed mutagenesis of key residues.

Fatal case of hemolytic-uremic syndrome in an adult due to a rare serogroup O91 Entero hemorrhagic Escherichia coli associated with a Clostridium difficile infection. More than meets the eye

open access: yesInternational Journal of Infectious Diseases, 2015
Hemolytic-uremic syndrome due to enterohemorrhagic Escherichia coli, belonging to serogroup O91 has rarely been described. We report here a case of post-diarrheal HUS due to EHEC O91 in an elderly patient for whom diagnosis was delayed given a previously

Production of antibacterial compounds by a Steely hybrid polyketide synthase in Dictyostelium

open access: yesFEBS Open Bio, EarlyView.
By selecting appropriate production species under optimised culture conditions, new chlorinated dibenzofuran compounds, which previously could not be analysed in sufficient quantities, were identified from cellular slime mould fruiting bodies. The strong antibacterial activity exhibited by these compounds suggests a link to the ecological function of ...

A general model for analysis of linear and hyperbolic enzyme inhibition mechanisms

open access: yesFEBS Open Bio, EarlyView.
We developed a general enzyme kinetic model that integrates these six basic inhibition mechanism onto a single one. From this model, we deduced a general enzyme kinetic equation that through modulation of simple parameters, γ (the relative inhibitor affinity for two binding sites) and β (the reactivity of the enzyme–substrate–inhibitor complex), is ...

Cloning, expression, purification, and structural modeling of the Chandipura virus matrix protein

open access: yesFEBS Open Bio, EarlyView.
This research protocol offers a guide for the cloning, expression, and purification of the Chandipura virus matrix protein using E. coli. It also includes a step‐by‐step procedure for cloning, expressing, and conducting fluorescence imaging of GFP‐fused Chandipura virus matrix protein in mammalian cell lines.

Homologous expression and purification of human HAX‐1 for structural studies

open access: yesFEBS Open Bio, EarlyView.
This research protocol provides detailed instructions for cloning, expressing, and purifying large quantities of the intrinsically disordered human HAX‐1 protein, N‐terminally fused to a cleavable superfolder GFP, from mammalian cells. HAX‐1 is predicted to undergo posttranslational modifications and to interact with membranes, various cellular ...

Computational Health Engineering Applied to Model Infectious Diseases and Antimicrobial Resistance Spread

open access: yesApplied Sciences, 2019
Infectious diseases are the primary cause of mortality worldwide. The dangers of infectious disease are compounded with antimicrobial resistance, which remains the greatest concern for human health.

ATP-Dependent Persister Formation in Escherichia coli

open access: yesmBio, 2017
Persisters are dormant variants that form a subpopulation of cells tolerant to antibiotics. Persisters are largely responsible for the recalcitrance of chronic infections to therapy.

Research progress on the metabolic transformation of Shenling Baizhu San under the influence of the gut microbiota

open access: yesAdvanced Chinese Medicine, EarlyView.
Abstract The gut microbiota plays a pivotal role in maintaining host metabolic homeostasis and overall physiological functions. Shenling Baizhu San (SLBZS), a time‐honored classical prescription for tonifying Qi, strengthening the spleen, removing dampness, and arresting diarrhea, has shown broad clinical efficacy in digestive disorders.
