Solid phase capture method for the specific amplification of microbial nucleic acids—avoidance of false-positive and false-negative reactions

open access: yesMolecular and Cellular Probes, 1991
DNA amplification assays such as the polymerase chain reaction are being developed for the amplification of small quantities of microbial nucleic acids. These assays offer the potential for a great deal of sensitivity. However, the high level of sensitivity increases the likelihood of cross-contamination of amplified products and the generation of ...

Adaptor protein CIN85 potentiates the motility of osteosarcoma cells via the Akt/mTOR and MMP2‐COL3A1 axis

open access: yesMolecular Oncology, EarlyView.
CIN85 is highly expressed in osteosarcoma, particularly in metastatic lesions. Its overexpression increases cell migration and Matrigel invasion, while silencing CIN85 suppresses these behaviors. Transcriptome analysis shows that CIN85 regulates MMP2, COL3A1, and Akt/mTOR signaling. Targeting these pathways reverses CIN85‐induced motility, highlighting

PECULIARITIES OF CONTROL OF HUMAN ALBUMIN DRUGS ON THE CONTENT OF BACTERIAL ENDOTOXINS

open access: yesРазработка и регистрация лекарственных средств, 2019
The problem was considered to determine bacterial endotoxins in human albumin drugs. To improve the safety of medicines the option of dual control was recommended, which includes the rabbit pyrogen test and determining bacterial endotoxins by the LAL ...

REGULATORY AND METHODOLOGICAL ASPECTS OF STUDYING ALLERGENIC PROPERTIES OF NEW MEDICINES AT THE PRECLINICAL STAGE

open access: yesРегуляторные исследования и экспертиза лекарственных средств, 2018
The assessment of allergic risk at the preclinical stage of drug development has been a debatable issue due to the integration of national requirements for medicines authorisation with those of the Eurasian Economic Union (EAEU).

Association between False Positive, False Negative, High-Background Cases and Humidity in One Lambda LABTypeTM HLA-DR Typing

open access: yesKorean Journal of Clinical Laboratory Science, 2023
Human leukocyte antigen (HLA) typing was performed in the diagnostic immunology laboratory of the Seoul National University Hospital. Among 611 HLA-DR tests, specific bead reactions suspected of being false positive and false negative in Lot 20 reagents ...

False-Positive Clostridium difficile in Negative-Control Reactions Peak and Then Decrease with Repetitive Refrigeration of Immunoassay [PDF]

open access: yesInternational Scholarly Research Notices, 2014
Aberrant false-positive reactions in negative-controls during ELISA testing for Clostridium difficile indicated the potential for false-diagnoses. Experiments with 96-well products showed a maximum peak of false-positive immunoassay reactions with the provided negative-control reagents after 5 refrigeration-to-room temperature cycles (P<0.001 ...

Longitudinal circulating tumor DNA profiling in patients with advanced endometrial cancer using an off‐the‐shelf targeted NGS panel

open access: yesMolecular Oncology, EarlyView.
Intratumour heterogeneity complicates precision management of advanced endometrial cancer. Circulating tumor DNA (ctDNA) offers a minimally invasive strategy to capture tumor evolution and therapeutic resistance. Here, we compare tumor‐agnostic NGS with tumor‐informed ddPCR, outlining their relative sensitivity, concordance, and clinical implications ...

Primary Immune Response Provides Protective Efficacy against SARS-CoV-2 Reinfection

open access: yesJournal of Nepal Medical Association, 2021
While there is absolutely no evidence to ensure recovered patients are either likely or unlikely to get reinfected. But studies in non-human primates indicate that reinfection of recovered patients is highly unlikely. It is also clear that primary immune

DNA methylation and expression of MAPRE3 affect overall survival of early‐stage non‐small cell lung cancer patients

open access: yesMolecular Oncology, EarlyView.
Both cg12821679MAPRE3 methylation and MAPRE3 expression are significantly associated with overall survival (OS) of non‐small cell lung cancer. Meanwhile, MAPRE3 expression significantly modified the effect of smoking cessation on OS. Smoking cessation benefits OS merely for patients with high MAPRE3 expression.

Validation and evaluation of RT-PCR real time in house test to detection of SARS-CoV-2 using specific RdRp gene and GAPDH endogen control

open access: yesRevista Peruana de Medicina Experimental y Salud Pública, 2021
The present work validated and evaluated a duplex real-time RT-PCR using specific primers and probes for genes RdRp from SARS-CoV-2 and GAPDH from humans; the latter was used as an endogenous control in all reactions.
