Comparison of Fed-batch Strategies in Hybridoma Cultures

2008
The classical cell culture process in stirred tank or airlift reactors is still the mostly used large scale production system. Fed-batch processes with different feeding strategies have been investigated to improve the productivity of hybridoma cultures.

Robust Control of Yeast Fed-Batch Cultures for Productivity Enhancement

IFAC Proceedings Volumes, 2009
Abstract Abstract This work proposes a robust control strategy for the optimizing control of fed-batch cultures of S. cerevisae. The process dynamics is characterized by a nonlinear kinetic model based on the bottleneck assumption and ethanol inhibition for a possible excess of substrate feeding.

Improvement of mammalian cell fed-batch culture

1995
Typical features of mammalian cells grown in suspension culture are the limitation of growth and very often the initiation of the cell death phase immediately after a critical cell density has been achieved. However, both features can be influenced to a certain extent by fortifying the growth medium as well as by feeding the culture with nutrients ...

Regulation of glutamine synthetase in fed-batch cultures of Neurospora crassa

Biochemical and Biophysical Research Communications, 1977
Abstract The effect of the nitrogen and carbon sources in the regulation of g l u tamine synthetase has been studied in fed-batch cultures of Neurospora crassa . The limitation of ammonium in an excess of the carbon source, leads to an accumulation of α-ketoglutarate and elevation of glutamine sy n thetase.

Fermentation kinetics of recombinant yeast in batch and fed‐batch cultures

Biotechnology and Bioengineering, 1992
AbstractFed‐batch cultures of recombinant microorganisms have attracted attention as they can separate cell growth stage from cloned‐gene expression phase during fermentations. In this work, the effect of different glucose feeding strategies on cell growth and cloned gene expression was studied during aerobic fed‐batch fermentations of recombinant ...

Production of the macrolide antibiotic tylosin in cyclic fed‐batch culture

Biotechnology and Bioengineering, 1987
AbstractTylosin‐producing Streptomyces fradiae was cultured on a synthetic medium with a high glutamate–glucose ratio. Tylosin batch fermentations with this medium were characterized by a high initial specific production rate of tylosin (qtylosin, mg/g h) that decreased as the fermentation progressed.

Comparison of optimization methods for fed-batch cultures of hybridoma cells

Bioprocess Engineering, 1997
Two methods for the calculation of optimal trajectories for the input variables of a fed-batch culture of hybridoma cells are compared. It pointed out that a gradient method based on Pontryagins' minimum principle based yields a significant better performance with respect to computational effort and the calculated minimum than a dynamic programming ...

The principle of parabolic feed for a fed-batch culture of baker’s yeast

Applied Biochemistry and Biotechnology, 1993
A parabolic nutrient supplementation has been derived by the specific growth rate, sugar conversion rate, and Pasteur effect, followed by high qualitative and quantitative biomass production with the lowest sugar consumption. The method produced about 95% cells in the G1 phase that were more resistant to drying and aging.

Growth of Photorhabdus luminescens in batch and glucose fed-batch culture

Applied Microbiology and Biotechnology, 2000
Photorhabdus luminescens, a bacterial symbiont of entomopathogenic biocontrol nematodes, was grown in batch and glucose fed-batch culture. The cell density, bioluminescence, production of antibiotic substances, number of cells with inclusion bodies, glucose concentration and oxygen uptake rate were recorded.

Comparison of a quadroma and its parent hybridomas in fed batch culture

Journal of Biotechnology, 1993
A quadroma (#22 x 63), formed by the fusion of two hybridomas, and its parent hybridomas (#22 and FMC 63) were each grown in fed batch cultures in order to examine the change in antibody productivity over time of the quadroma compared to its parent hybridomas.
