Etoposide‐induced cancer cell death: roles of mitochondrial VDAC1 and calpain, and resistance mechanisms

open access: yesMolecular Oncology, EarlyView.
The complex mode of action of the topoisomerase II inhibitor etoposide in triggering apoptosis involves several mechanisms: overexpression of the mitochondrial protein VDAC1, leading to its oligomerization and formation of a large channel that mediates the release of pro‐apoptotic protein; and overexpression of the apoptosis regulators p53, Bax, and ...

Peripheral blood leukocyte signatures as biomarkers in relapsed ovarian cancer patients receiving combined anti‐CD73/anti‐PD‐L1 immunotherapy in arm A of the NSGO‐OV‐UMB1/ENGOT‐OV30 trial

open access: yesMolecular Oncology, EarlyView.
Using mass cytometry, we analyzed serial blood samples from patients with relapsed epithelial ovarian cancer (EOC) treated with oleclumab–durvalumab combination immunotherapy in the NSGO‐OV‐UMB1/ENGOT‐OV30 trial. Our analysis identified potential predictive, monitoring, and response biomarkers detectable through liquid biopsy. These findings facilitate

Response to neoadjuvant chemotherapy in early breast cancers is associated with epithelial–mesenchymal transition and tumor‐infiltrating lymphocytes

open access: yesMolecular Oncology, EarlyView.
Epithelial–mesenchymal transition (EMT) and tumor‐infiltrating lymphocytes (TILs) are associated with early breast cancer response to neoadjuvant chemotherapy (NAC). This study evaluated EMT and TIL shifts, with immunofluorescence and RNA sequencing, at diagnosis and in residual tumors as potential biomarkers associated with treatment response.

Investigations of dynamic light scattering properties in fluorescent solution: A comprehensive study using plasmonic enhanced Flow-Cytometry [PDF]

open access: yesarXiv, 2017
In this article we present a new diagnostic approach utilizing flow-cytometry to study compounds of nanoparticle samples in solution by analysis of their scattering patterns retrieved from the cytometric measurements. As a specific enhancement of this technique we study as well the scattering pattern of nanoparticles in a fluorescent solution (529 nm).

Multiplex single‐cell profiling of putative cancer stem cell markers ALDH1, SOX9, SOX2, CD44, CD133 and CD15 in endometrial cancer

open access: yesMolecular Oncology, EarlyView.
Cancer stem cells are associated with aggressive disease, but a deep characterization of such markers is lacking in endometrial cancer. This study uses imaging mass cytometry to explore putative cancer stem cell markers in endometrial tumors and corresponding organoid models.

Flow Cytometry as a Diagnostic Tool in the Early Diagnosis of Aggressive Lymphomas Mimicking Life-Threatening Infection

open access: yesCase Reports in Medicine, 2011
Aggressive lymphomas can present with symptoms mimicking life-threatening infection. Flow cytometry (FC) is usually recommended for the classification and staging of lymphomas in patients with organomegaly and atypical cells in effusions and blood, after

An Algorithmic Pipeline for Analyzing Multi-parametric Flow Cytometry Data [PDF]

open access: yesarXiv, 2015
Flow cytometry (FC) is a single-cell profiling platform for measuring the phenotypes of individual cells from millions of cells in biological samples. FC employs high-throughput technologies and generates high-dimensional data, and hence algorithms for analyzing the data represent a bottleneck.

Peripheral blood proteome biomarkers distinguish immunosuppressive features of cancer progression

open access: yesMolecular Oncology, EarlyView.
Immune status significantly influences cancer progression. This study used plasma proteomics to analyze benign 67NR and malignant 4T1 breast tumor models at early and late tumor stages. Immune‐related proteins–osteopontin (Spp1), lactotransferrin (Ltf), calreticulin (Calr) and peroxiredoxin 2 (Prdx2)–were associated with systemic myeloid‐derived ...
