Regulatory T-Cells, FoxP3 and Atherosclerosis
2009Innate immune responses follow accumulation of modified lipids within the arterial wall thereby influencing atherosclerotic plaque progression. One of the mechanisms evolved in maintaining immunologic self-tolerance involves upregulation of regulatory T-cells, among which the CD4+CD25+ FoxP3+ regulatory T-cells (Treg) are best characterized.

Immune Regulation Although expression of FoxP3 is largely synonymous with T regulatory (Treg) cell identity in mice, type 1 regulatory T cells (TR1) are an exception. TR1 cells produce interleukin-10 but are FoxP3-negative. In comparison with FoxP3-positive Treg cells, the development and functions of TR1 cells are poorly understood.

Some regulatory T cells express the Foxp3 transcription factor and such Tregs have an essential function of preventing autoimmune disease in man and mouse. Foxp3 binds to Forkhead motifs of about 1100 genes and the strength of binding increases when Foxp3-expressing T cells are stimulated by PMA and ionomycin.
