Generating golden Syrian hamsters with conditional alleles via zygote microinjection of CRISPR/Cas9

open access: yesAnimal Models and Experimental Medicine, EarlyView.
We established the first conditional knockout (cKO) model in the golden Syrian hamster by CRISPR/Cas9‐mediated genome editing. Cas9 protein, two sgRNAs, and a donor plasmid carrying loxP‐flanked exon 2 of the ApoF gene were microinjected into one‐cell embryos. The floxed allele was efficiently generated (up to 27%) and transmitted through the germline.

High Dietary Fructose: Direct or Indirect Dangerous Factors Disturbing Tissue and Organ Functions

open access: yesNutrients, 2017
High dietary fructose is a major contributor to insulin resistance and metabolic syndrome, disturbing tissue and organ functions. Fructose is mainly absorbed into systemic circulation by glucose transporter 2 (GLUT2) and GLUT5, and metabolized in liver ...

Chronic high‐fat diet induces multi‐organ dysfunction and metabolic homeostasis disruption in Macaca fascicularis

open access: yesAnimal Models and Experimental Medicine, EarlyView.
An 18‐month HFD successfully established a translational Macaca fascicularis model replicating key metabolic disorders (MASH, diabetes, cardiac hypertrophy). MASH was determined by liver biopsy histology, the presence steatosis, inflammatory infiltration, hepatocytic ballooning, and fibrosis were considered as MASH; diabetes was diagnosed according to ...

Elasticity sampling links thermodynamics to metabolic control

open access: yes, 2013
Metabolic networks can be turned into kinetic models in a predefined steady state by sampling the reaction elasticities in this state. Elasticities for many reversible rate laws can be computed from the reaction Gibbs free energies, which are determined ...

Formation of Fructose-Mediated Advanced Glycation End Products and Their Roles in Metabolic and Inflammatory Diseases.

open access: yesAdvances in Nutrition, 2017
Fructose is associated with the biochemical alterations that promote the development of metabolic syndrome (MetS), nonalcoholic fatty liver disease, and type 2 diabetes. Its consumption has increased in parallel with MetS. It is metabolized by the liver,

Establishment of a novel cynomolgus monkey model of hyperuricemia

open access: yesAnimal Models and Experimental Medicine, EarlyView.
We established a novel hyperuricemia model in cynomolgus monkeys (A) via oral administration of potassium oxonate, hydrochlorothiazide, and adenine mixed in fruits and vegetables. This model (B) targets uric acid synthesis and enzymatic degradation, and (C) regulates renal and intestinal excretion, thereby stably elevating serum uric acid and serving ...

Rat brain serotonin synthesis under prolonged consumption of fructose solution

open access: yesБіологічні студії, 2014
Key metabolites and enzymes of rat brain serotonin biosynthesis pathway under the development of obesity induced by the consumption of 10% fructose solution, have been studied. A reduction of tryptophan by 60±10 % (p < 0.05, n = 10), 5-hydroxytryptophan,

The colorimetric determination of hexoses with carbazole [PDF]

open access: yes, 1947
The optimum conditions for the calorimetric estimation of hexoses by reaction with carbazole in hot sulfuric acid solution have been determined and a convenient procedure, giving results with a precision of 2 to 5 per cent in the range of 50 to 150 γ of ...

Uric acid activates aldose reductase and the polyol pathway for endogenous fructose and fat production causing development of fatty liver in rats

open access: yesJournal of Biological Chemistry, 2019
Dietary, fructose-containing sugars have been strongly associated with the development of nonalcoholic fatty liver disease (NAFLD). Recent studies suggest that fructose also can be produced via the polyol pathway in the liver, where it may induce hepatic
