Versatile synthesis of amino acid functionalized nucleosides via a domino carboxamidation reaction

open access: yesBeilstein Journal of Organic Chemistry, 2014
Functionalized oligonucleotides have recently gained increased attention for incorporation in modified nucleic acid structures both for the design of aptamers with enhanced binding properties as well as the construction of catalytic DNA and RNA.

A comparison of synthetic oligodeoxynucleotides, DNA fragments and AAV-1 for targeted episomal and chromosomal gene repair

open access: yesBMC Biotechnology, 2009
Background Current strategies for gene therapy of inherited diseases consist in adding functional copies of the gene that is defective. An attractive alternative to these approaches would be to correct the endogenous mutated gene in the affected ...

Enhancing Small Molecule Sensing With Aptameric Functionalized Nano Devices

open access: yesAdvanced Materials Technologies, EarlyView.
Unveiling an ultra‐sensitive, non‐invasive neurotransmitter sensor. For the first time, a nanoscale sensor for detecting an important neurotransmitter was demonstrated using micro‐electromechanical systems (MEMS) technology. Our approach utilized field‐effect transistor (FET)‐based readout to enable pico‐molar detection of biomarkers in sweat.

Targeting splicing for hematological malignancies therapy

open access: yesBMC Genomics
Alterations in splicing patterns of leukemic cells have a functional impact and influence most cellular processes since aberrantly spliced isoforms can provide a proliferative advantage, enable to evade apoptosis, induce metabolic reprogramming, change ...

Cell Cycle Control of Nuclear Metabolism Couples Phosphatidylinositol Signaling to Histone Methylation

open access: yesAdvanced Science, EarlyView.
Nuclear metabolism oscillates during cell cycle progression. Quantitative chromatome proteomics and imaging reveal phase‐specific dynamics of PIP5K1A and nuclear PIP2, linking phosphatidylinositol metabolism to histone methylation. This work identifies nuclear lipid metabolism as a previously unrecognized regulatory axis coordinating chromatin ...

Tailoring Vascular‐Immune Homeostasis via Manganese‐DNA Complex‐Armed Immunogenic Extracellular Vesicles for Pancreatic Cancer Immunotherapy

open access: yesAdvanced Science, EarlyView.
This study demonstrates that Mn2⁺–tumor DNA complexes encapsulated in dendritic cell (DC)– derived immunogenic extracellular vesicles (EVDC@Mn‐DNA) act as a DC‐specific cGAS– STING activator. EVDC@Mn‐DNA treatment enhances intratumoral DC activation, improves tumor vascular function, promotes CD8⁺ T cell activity, and suppresses pancreatic tumor growth,

miR‐33 controls the expression of biliary transporters, and mediates statin‐ and diet‐induced hepatotoxicity

open access: yesEMBO Molecular Medicine, 2012
Bile secretion is essential for whole body sterol homeostasis. Loss‐of‐function mutations in specific canalicular transporters in the hepatocyte disrupt bile flow and result in cholestasis.

Correlating In Vitro Splice Switching Activity With Systemic In Vivo Delivery Using Novel ZEN-modified Oligonucleotides

open access: yesMolecular Therapy: Nucleic Acids, 2014
Splice switching oligonucleotides (SSOs) induce alternative splicing of pre-mRNA and typically employ chemical modifications to increase nuclease resistance and binding affinity to target pre-mRNA. Here we describe a new SSO non-base modifier (a naphthyl-
