Structural insights into an engineered feruloyl esterase with improved MHET degrading properties

open access: yesFEBS Letters, EarlyView.
A feruloyl esterase was engineered to mimic key features of MHETase, enhancing the degradation of PET oligomers. Structural and computational analysis reveal how a point mutation stabilizes the active site and reshapes the binding cleft, expading substrate scope.

A methionine‐lined active site governs carbocation stabilization and product specificity in a bacterial terpene synthase

open access: yesFEBS Letters, EarlyView.
This study reveals a unique active site enriched in methionine residues and demonstrates that these residues play a critical role by stabilizing carbocation intermediates through novel sulfur–cation interactions. Structure‐guided mutagenesis further revealed variants with significantly altered product profiles, enhancing pseudopterosin formation. These

A quasi-3D hyperbolic shear deformation theory for functionally graded plates [PDF]

open access: yes, 2013
A quasi-3D hyperbolic shear deformation theory for functionally graded plates is developed. The theory accounts for both shear deformation and thickness-stretching effects by a hyperbolic variation of all displacements across the thickness, and satisfies

Aggressive prostate cancer is associated with pericyte dysfunction

open access: yesMolecular Oncology, EarlyView.
Tumor‐produced TGF‐β drives pericyte dysfunction in prostate cancer. This dysfunction is characterized by downregulation of some canonical pericyte markers (i.e., DES, CSPG4, and ACTA2) while maintaining the expression of others (i.e., PDGFRB, NOTCH3, and RGS5).

Isogeometric free vibration of sector cylindrical shells with carbon nanotubes reinforced and functionally graded materials

open access: yesResults in Physics, 2020
An isogeometric numerical procedure based on non-uniform rational B-splines is applied to solve the free vibration of carbon nanotubes reinforced and functionally graded material sector cylindrical shells.

Cytoplasmic p21 promotes stemness of colon cancer cells via activation of the NFκB pathway

open access: yesMolecular Oncology, EarlyView.
Cytoplasmic p21 promotes colorectal cancer stem cell (CSC) features by destabilizing the NFκB–IκB complex, activating NFκB signaling, and upregulating BCL‐xL and COX2. In contrast to nuclear p21, cytoplasmic p21 enhances spheroid formation and stemness transcription factor CD133.

Fabrication of Functionally Graded Material (FGM) Set-Up [PDF]

open access: yes, 2015
Functionally graded materials (FGMs) are advanced composites with graded properties that are novel concepts in the structural design of materials and have extensive applications in the aerospace engineering, aircrafts and automotive industry.

In vitro models of cancer‐associated fibroblast heterogeneity uncover subtype‐specific effects of CRISPR perturbations

open access: yesMolecular Oncology, EarlyView.
Development of therapies targeting cancer‐associated fibroblasts (CAFs) necessitates preclinical model systems that faithfully represent CAF–tumor biology. We established an in vitro coculture system of patient‐derived pancreatic CAFs and tumor cell lines and demonstrated its recapitulation of primary CAF–tumor biology with single‐cell transcriptomics ...

State-space Levy solution for size-dependent static, free vibration and buckling behaviours of functionally graded sandwich plates [PDF]

open access: yes, 2018
The size-dependent static, free vibration and buckling behaviours of functionally graded (FG) sandwich plates are analysed in this study. Utilising the modified couple stress theory and variational principle, governing equations of motion are developed ...

Transcriptional network analysis of PTEN‐protein‐deficient prostate tumors reveals robust stromal reprogramming and signs of senescent paracrine communication

open access: yesMolecular Oncology, EarlyView.
Combining PTEN protein assessment and transcriptomic profiling of prostate tumors, we uncovered a network enriched in senescence and extracellular matrix (ECM) programs associated with PTEN loss and conserved in a mouse model. We show that PTEN‐deficient cells trigger paracrine remodeling of the surrounding stroma and this information could help ...
