Identification and characterization of a homozygous deletion found in ovarian ascites by representational difference analysis

We have performed representational difference analysis (RDA) on DNA from tumor cells and normal fibroblasts isolated from the ascites of a patient with ovarian cancer. Five of six products of the RDA were homozygously deleted from the tumor DNA.

Targeting a Myeloid–Regulatory B Cell Network Reverses Immune Paralysis in Periprosthetic Joint Infections

Periprosthetic joint infection establishes a sophisticated immunosuppressive network between CXCR4+ PMN‐MDSCs and Bregs, inducing profound CD8+ T cell paralysis. Alendronate effectively disrupts this CXCR4+ PMN‐MDSC–Breg axis by targeting STAT3, thereby restoring local immune surveillance.

Matrix Stiffness Induces Endothelial Network Senescence

Using a 3D human in vitro model that decouples mechanical stress from inflammatory or biochemical signals, matrix stiffening induces a senescence phenotype in endothelial networks. This mechano‐induced senescence activates Notch signaling, and pharmacologic Notch inhibition attenuates this stiffness‐induced senescence.
