Mechanism of glucocorticoid receptor down-regulation by glucocorticoids.

The effect of glucocorticoids on the regulation of glucocorticoid receptor mRNA was studied in two different cell lines, human IM-9 lymphocytes and rat pancreatic acinar AR42J cells. Using a glucocorticoid receptor cDNA probe, glucocorticoid receptor mRNA was examined by Northern blot hybridization and quantitated by slot-blot hybridization.

Rictor Ameliorates Acute Antibody‐Mediated Rejection Following Kidney Transplantation by Suppressing Macrophage M1 Polarization Through p65‐NLRP3 Axis

This study elucidates the critical role of Rictor in macrophage activation in acute antibody‐mediated rejection (ABMR). Rictor increases K48‐linked ubiquitination of p65 by upregulating E3 ubiquitin ligase SOCS1, inhibiting transcriptional levels of NLRP3 and inflammasome activation.

Single‐cell transcriptomics and genetic risk mapping revealed a shift from LCN2high myeloid‐derived suppressor cells maintaining disc repair in early stages to IL1B+ macrophage‐dominated pathology in advanced disease.
