Current Opinion in Chemical Biology, 2000
Insights into glycosidase mechanisms have come from X-ray crystallographic studies on complexes with substrate analogs and inhibitors, representing all the intermediate species along the reaction coordinate. Site-directed mutagenesis continues to play a significant role in understanding mechanisms, but is also proving important in generating ...

Current Opinion in Chemical Biology, 2002
The three-dimensional structure of glycosidases and of their complexes and the study of transition-state mimics reveal structural details that correlate with mechanism. Of particular interest are the transition-state conformations harnessed by individual enzymes and the substrate distortion observed in enzyme-ligand complexes.

From Covalent Glycosidase Inhibitors to Activity‐Based Glycosidase Probes
Chemistry – A European Journal, 2014AbstractActivity‐based protein profiling has emerged as a powerful discovery tool in chemical biology and medicinal chemistry research. Success of activity‐based protein profiling hinges on the presence of compounds that can covalently and irreversibly bind to enzymes, do so selectively in the context of complex biological samples, and subsequently ...

A NUMBER of sources of plant glycosidases are known, notably almonds and lucerne seed1. Having suitable substrates available, we have made a survey of a large number of common garden, farm and wayside plants for different glycosidase activities.

Annual Review of Biochemistry, 1999
▪ Abstract Enzymatic hydrolysis of glycosides can occur by one of two elementary mechanisms identified by the stereochemical outcome of the reaction, inversion or retention. The key active-site residues involved are a pair of carboxylic acids in each case, and strategies for their identification and for probing the details of their roles in catalysis

Diglycoside-Specific Glycosidases
2003Publisher Summary The chapter focuses on β -primeverosidase, which is is one of the disaccharide-specific glycosidases, or “diglycosidases.” A β -primeverosidase responsible for the formation of aroma from the glycosidic aroma precursors was purified from the crude enzyme extract prepared from fresh juvenile tea leaves (Camellia sinensis).

Glycosidase Activities of Mycoplasmas
Zentralblatt für Bakteriologie, 1990The activities of alpha- and beta-glucosidase, beta-galactosidase and beta-N acetylglucosaminidase were assessed at acidic pH by fluorimetry using the appropriate 4-methylumbelliferyl substrate in four Mycoplasma species (M. pneumoniae, M. gallisepticum, M. hominis and M. capricolum) and in Acholeplasma laidlawii.

Glycosidases ofTrichinella spiralis
Molecular and Biochemical Parasitology, 1985The exoglycosidases beta-N-acetyl-D-glucosaminidase, beta-N-acetyl-D-galactosaminidase, alpha-1-fucosidase, alpha-D-glucosidase and alpha-D-mannosidase, and a non-specific acid phosphohydrolase are present at high levels in extracts of adult and muscle-stage (L1) Trichinella spiralis and at lower (5-30-fold) levels in extracts of the newborn larvae ...
