Phospholipid hydroperoxide glutathione peroxidase (PHGPx), the product of gpx-4, is the major selenoprotein in sperm and is considered essential for fertilization because of its multiple roles in spermatogenesis, such as hydroperoxide detoxification, formation of the mitochondrial capsule, and chromatin condensation.

Insight from GPxs structure and function

The multigene family of GPxs encodes for tetrameric and monomeric enzymes both containing either a selenocysteine (Sec) or cysteine (Cys) residue at the catalytic site. Monomeric GPxs carry deletions of the subunit interfaces. The Sec containing homologues are found in vertebrates, and, with a scattered presence, in lower organisms.

Efficacy of a killed gpX deleted pseudorabies virus vaccine.

Ten inactivated vaccines containing one of four adjuvants and varying concentrations of pseudorabies virus (PRV) antigens were compared in order to select a vaccine suitable for commercial production. A genetically engineered strain of PRV lacking the gene coding for glycoprotein X (gpX) was used in these vaccines.
