Glutathione peroxidase 4 (GPX4) and obesity interact to impact tumor progression and treatment response in triple negative breast cancer

open access: yesCancer & Metabolism
Introduction Triple-negative breast cancer (TNBC), which tends to be more advanced when diagnosed and more aggressive than other breast cancer subtypes, is accelerated by obesity.

Human mesenchymal stem cells derived exosomes improve ovarian function in chemotherapy-induced premature ovarian insufficiency mice by inhibiting ferroptosis through Nrf2/GPX4 pathway

open access: yesJournal of Ovarian Research
Chemotherapy exposure has become a main cause of premature ovarian insufficiency (POI). This study aimed to evaluate the role and molecular mechanism of human umbilical cord mesenchymal stem cell-derived exosomes (hUMSC-Exos) in ovarian function ...

Targeting Peptostreptococcus anaerobius with an Iron‐Based Nanozyme Reverses Ferroptosis Resistance and Enhances Antitumor Immunity in Colorectal Cancer

open access: yesAdvanced Science, EarlyView.
An iron‐based nanozyme selectively eliminates intratumoral P. anaerobius while catalytically generating ROS to induce ferroptosis, synergistically suppressing colorectal cancer growth and activating anti‐tumor immunity through immunogenic cell death. ABSTRACT The intratumoral microbiota is a critical determinant of therapeutic outcomes in colorectal ...

Mechanism of LncRNA NORAD regulating ferroptosis in endometrial cancer cells by modifying GPX4 through FTO-mediated m6A methylation

open access: yesCellular and Molecular Life Sciences
Objective Endometrial cancer (EC) represents the most prevalent gynecologic malignancy. We probed the mechanism of long noncoding RNA activated by DNA damage (lncRNA NORAD) regulating ferroptosis in EC cells (ECCs) by modifying glutathione peroxidase 4 ...

Pirin Transcriptionally Regulates PLA2G4A To Inhibit Ferroptosis in Colorectal Cancer via Lipid Profile Remodeling

open access: yesAdvanced Science, EarlyView.
This study identifies Pirin (PIR), an iron‐binding protein, as a critical ferroptosis suppressor in colorectal cancer through lipid membrane remodeling. PIR, induced by NRF2 during ferroptotic stress, transcriptionally regulates PLA2G4A to shift cellular lipid composition away from ferroptosis‐permissive polyunsaturated phospholipids.

The role of glutathione peroxidase 4 in neuronal ferroptosis and its therapeutic potential in ischemic and hemorrhagic stroke

open access: yesBrain Research Bulletin
Ferroptosis is a type of cell death that depends on iron and is driven by lipid peroxidation, playing a crucial role in neuronal death during stroke. A central element in this process is the inactivation of glutathione peroxidase 4 (GPx4), an antioxidant

HMGA2 alleviates ferroptosis by promoting GPX4 expression in pancreatic cancer cells

open access: yesCell Death and Disease
Pancreatic cancer is one of the most malignant tumor types and is characterized by high metastasis ability and a low survival rate. As a chromatin-binding protein, HMGA2 is widely overexpressed and considered an oncogene with various undefined regulatory

CRISPLD2 Attenuates Intervertebral Disc Degeneration by Suppressing Oxidative Stress‐Induced Ferroptosis through the miR‐548I‐IL17A Axis

open access: yesAdvanced Science, EarlyView.
This study identifies CRISPLD2 as a key protector against IVDD. By regulating ferroptosis through the CRISPLD2–miR‐548I–IL17A axis, CRISPLD2 maintains NPCs homeostasis and reduces oxidative stress. Restoring CRISPLD2 expression effectively alleviates disc degeneration and highlights a promising therapeutic strategy for discogenic low back pain ...
