Structure‐Guided Engineering of a Cas12i Nuclease Unlocks Near‐PAMless Genome Editing

open access: yesAdvanced Science, EarlyView.
CRISPR‐Cas nucleases are limited by PAM requirements, restricting genome accessibility. Structure‐guided engineering of the compact Cas12i nuclease SF01 produced three variants with near‐PAMless, enabling efficient editing at diverse 5'‐NNTN‐3' sites. These nucleases expand the editable portion of the human genome more than fourfold, enabling efficient

Characterization and bioinformatic filtering of ambient gRNAs in single-cell CRISPR screens using CLEANSER

open access: yesCell Genomics
Summary: Single-cell RNA sequencing CRISPR (perturb-seq) screens enable high-throughput investigation of the genome, allowing for characterization of thousands of genomic perturbations on gene expression. Ambient gRNAs, which are contaminating gRNAs, are

Enhanced CRISPR/Cas9-mediated precise genome editing by improved design and delivery of gRNA, Cas9 nuclease, and donor DNA.

open access: yesJournal of Biotechnology, 2017
While CRISPR-based gene knock out in mammalian cells has proven to be very efficient, precise insertion of genetic elements via the cellular homology directed repair (HDR) pathway remains a rate-limiting step to seamless genome editing.

Metabolic gatekeeper function of B-lymphoid transcription factors. [PDF]

open access: yes, 2017
B-lymphoid transcription factors, such as PAX5 and IKZF1, are critical for early B-cell development, yet lesions of the genes encoding these transcription factors occur in over 80% of cases of pre-B-cell acute lymphoblastic leukaemia (ALL).

Alphaviral Capsid Proteins Inhibit Stress Granule Assembly via Competitive RNA Binding With G3BP1

open access: yesAdvanced Science, EarlyView.
Stress granules exert antiviral functions. This study illustrates a conserved function of alphaviral capsid proteins in modulating stress granules. Oligomerization mediated by a helical motif coupled with a positively charged intrinsically disordered region (IDR) directly competes with G3BP1 for RNA binding, thereby disrupting G3BP1‐RNA liquid–liquid ...

IGFBP5 Restores Endometrial Receptivity and Rescues Implantation Failure in Polycystic Ovary Syndrome

open access: yesAdvanced Science, EarlyView.
Impaired endometrial receptivity contributes to the poor pregnancy outcomes in women with PCOS. The authors found that decreased endometrial IL‐22 levels and disrupted STAT3‐IGFBP5 signaling pathway contributed to impaired endometrial receptivity. Supplementation with IL‐22 or IGFBP5 exerts a protective effect on implantation failure in PCOS‐like mice,

Determinants of crispr nuclease grna function

open access: yes, 2023
CRISPR (clustered regularly interspaced palindromic repeats) systems are sought-after genome editing tools for therapeutics and diagnostics. The use of CRISPR systems for gene editing is not without difficulties: the specificity of Cas12a nuclease, a Cas9 alternative, presents a limit in its use for gene editing due to its inability to successfully ...

Dysregulation of the PATZ1/CTCF Balance Silences ZBTB20 to Drive Melanoma Progression

open access: yesAdvanced Science, EarlyView.
This study uncovers a new oncogenic mechanism in melanoma. The transcription factor PATZ1 competes with the architectural protein CTCF for DNA binding, thereby disrupting a specific chromatin loop and silencing the tumor suppressor ZBTB20. This event unleashes the pro‐tumorigenic PMEPA1‐p38‐STAT1 signaling axis, promoting cancer progression.

Mismatch prime editing gRNA increased efficiency and reduced indels

open access: yesNature Communications
Prime editing enables precise and efficient genome editing, but its efficacy is hindered by pegRNA's 3' extension, forming secondary structures due to high complementarity with the protospacer. The continuous presence of the prime editing system also leads to unintended indel formation, raising safety concerns for therapeutic applications.

Pancancer Fine‐Mapping of Mutational Intolerance Identifies CHEK1 as an Immunosuppressive Driver in Lung Adenocarcinoma

open access: yesAdvanced Science, EarlyView.
This study identifies mutation‐intolerant genes (MIGs), which are mutationally constrained in tumors despite normal‐tissue variability. Using miDriver, the authors pinpoint MIGs essential for tumor‐intrinsic fitness and immune evasion. Focusing on CHEK1, they show it drives tumor fitness and sculpts an immunosuppressive niche via the MIF–CD74 axis ...
