Internal guide RNA interactions interfere with Cas9-mediated cleavage
While the CRISPR-Cas9 system has revolutionised molecular biology, it is still a mystery why not every guide RNA elicits target DNA cleavage. Here the authors show that genomic context and internal gRNA interactions can inhibit cleavage.

We achieved cytoplasmic delivery of non‐cell‐penetrating IgGs by grafting a single functional complementarity‐determining region 1 (CDR1) from the light chain variable region (VL) of the cell‐internalizable 3D8 antibody. The engineered IgG acquired cell‐penetrating ability while maintaining antigen affinity, highlighting CDR1 grafting as a promising ...

Prokaryotic Argonautes – variations on the RNA interference theme
The discovery of RNA interference (RNAi) has been a major scientific breakthrough. This RNA-guided RNA interference system plays a crucial role in a wide range of regulatory and defense mechanisms in eukaryotes.

Urine is a rich source of biomarkers for cancer detection. Tumor‐derived material is released into the bloodstream and transported to the urine. Urine can easily be collected from individuals, allowing non‐invasive cancer detection. This review discusses the rationale behind urine‐based cancer detection and its potential for cancer diagnostics ...

Generation of Mutant Pigs by Direct Pronuclear Microinjection of CRISPR/Cas9 Plasmid Vectors
A set of Cas9 and single guide CRISPR RNA expression vectors was constructed. Only a very simple procedure was needed to prepare specific single-guide RNA expression vectors with high target accuracy.

Subpar reporting of pre‐analytical variables in RNA‐focused blood plasma studies
Pre‐analytical variables strongly influence the analysis of extracellular RNA (cell‐free RNA; exRNA) derived from blood plasma. Their reporting is essential to allow interpretation and replication of results. By evaluating 200 exRNA studies, we pinpoint a lack of reporting pre‐analytical variables associated with blood collection, plasma preparation ...

Evolving Boolean Networks with RNA Editing [PDF]
The editing of transcribed RNA by other molecules such that the form of the final product differs from that specified in the corresponding DNA sequence is ubiquitous. This paper uses an abstract, tunable Boolean genetic regulatory network model to explore aspects of RNA editing.
