Natural Products Inspired Scaffold Diversification Leads to Unnatural Molecular Warhead and Covalent Strategy to Modulating Protein Function through Electrophilic Bromine Transfer

open access: yesAdvanced Science, EarlyView.
We report a new thiolate‐reactive α,α‐gem‐dibromo lactam warhead that activates transcription factor Nrf2 and demonstrates anti‐inflammatory activities, which have implications in cancer, neurodegeneration, and cardiovascular diseases. RNA‐seq illuminated detailed transcriptional profiles, and chemical reactions with cysteine‐containing compounds ...

Visualization of Endosomal Escape of Intracellularly Delivered Protein With Unexpected Photochemical Internalization of Trypan Blue

open access: yesAdvanced Science, EarlyView.
TB, a conventional cell viability dye, acts as both a fluorescence modulator (quenching GFP fluorescence upon protein binding and recovering it upon release) and a photosensitizer that triggers endosomal escape via photochemical internalization (PCI) under 590 nm light.

Simultaneous measurements of auto-immune and infectious disease specific antibodies using a high throughput multiplexing tool. [PDF]

open access: yesPLoS ONE, 2012
Considering importance of ganglioside antibodies as biomarkers in various immune-mediated neuropathies and neurological disorders, we developed a high throughput multiplexing tool for the assessment of gangliosides-specific antibodies based on Biolpex ...

Synthetic vaccine affords full protection to mice against lethal challenge of influenza B virus of both genetic lineages

open access: yesiScience, 2021
Summary: A quarter of all seasonal influenza cases are caused by type B influenza virus (IBV) that also dominates periodically. Here, we investigated a recombinant adenovirus vaccine carrying a synthetic HA2 representing the consensus sequence of all IBV

Origin of the Vaccinia Virus Hemagglutinin [PDF]

open access: yesJournal of Virology, 1972
The relationship between the vaccinia virus hemagglutinin and hemadsorption was examined. Hemagglutinin synthesis was temporally related to the appearance of the hemadsorption reaction. Only chicken erythrocytes, which reacted with hemagglutinin, hemadsorbed to infected cells, and both of these reactions were inhibited by Ca 2+

Lymphoid‐Tissue‐on‐Chip Recapitulates Human Antibody Responses In Vitro

open access: yesAdvanced Science, EarlyView.
The presented lymphoid‐tissue‐on‐chip system allows culture of primary human tonsil cells at organotypic high density under perfusion for up to 4 weeks, emulates immune response to soluble vaccines and vaccination via peripheral antigen‐presenting cells and represents a useful tool to assess cellular interactions during homeostasis, immune responses ...

Structural analysis of the variable major proteins of Borrelia hermsii. [PDF]

open access: yes, 1983
Borrelia hermsii undergoes spontaneous antigenic variation in vivo and in vitro. Serotype specificity is associated with expression of one of a family of molecular weight-variable proteins, the pI proteins.

Linker and/or transmembrane regions of influenza A/Group-1, A/Group-2, and type B virus hemagglutinins are packed differently within trimers.

open access: yesBiochimica et Biophysica Acta, 2011
Influenza virus hemagglutinin is a homotrimeric spike glycoprotein crucial for virions' attachment, membrane fusion, and assembly reactions. X-ray crystallography data are available for hemagglutinin ectodomains of various types/subtypes but not for ...

A Circuit of Mechanically Regulated Transcription Factors Balances Regenerative and Fibrotic Memory of Mesenchymal Stromal Cells

open access: yesAdvanced Science, EarlyView.
Producing MSCs on rigid culture substrates induces a scar‐making phenotype, jeapordizing therapeutic success. ‘Tissue‐soft’ surfaces prevent MSC fibrogenesis and preserve regenerative traits. An epigenetic network, driven by HOXA11 and SALL1, maintains ‘soft memory’ by keeping chromatin open in relaxed MSCs, promoting anti‐fibrotic programs.

Reduction in Post-Botulinum Toxin Flu-like Symptoms After Injection with Incobotulinum Toxin [PDF]

open access: yes, 2014
OBJECTIVE: To determine if patients reporting flu-like symptoms (FLS) after botulinum toxin (BoNT) injections are less susceptible to this reaction after incobotulinum toxin.
