A Comprehensive 19F NMR Framework for Fragment‐Based Drug Discovery: The Validated Screening Library OpenFL600 and Efficient Affinity Ranking by CSAR

open access: yesAngewandte Chemie, EarlyView.
NMR screening is a powerful method for hit detection in drug‐discovery. We designed and validated the OpenFL600 19F$^{19}{\rm F}$ NMR library to probe diverse targets, including RNA, GPCRs, kinases, and proteases. This library yields target‐specific ligands without generating promiscuous binders.

Zero‐Valent Nickel Stabilized in Surface‐Supported Metal–Organic Chains

open access: yesAngewandte Chemie International Edition, EarlyView.
9,10‐Dicyanoanthracene coordinates Ni atoms on Ag(100) to form metal–organic chains that stabilize zero‐valent Ni(0) (d10) centers with quasi‐one‐dimensional metal–ligand electronic coupling. ABSTRACT The stabilization of zero‐valent nickel Ni(0) centers in extended coordination networks remains a major challenge due to their pronounced reactivity and ...

Photogenerated Oxetanes as a Gateway to Uphill Cyclopropanation

open access: yesAngewandte Chemie International Edition, EarlyView.
An atom‐economical route to densely functionalized cyclopropanes from furans and photochemically activated aldehydes is enabled by a unique oxetane‐to‐cyclopropane rearrangement. These cyclopropanes can be used to access complex bioactive scaffolds and serve as a platform for asymmetric cyclopropane synthesis.

Minute‐Scale Repeated Metabolic Probing of Living Cells Enabled by Rapid Parahydrogen‐Based Hyperpolarization

open access: yesAngewandte Chemie International Edition, EarlyView.
Rapid parahydrogen hyperpolarization combined with simple dilution enables biocompatible delivery of hyperpolarized pyruvate to immobilized living cells within ≈1 min. Repeated substrate injections reveal frequency‐dependent metabolic dynamics, introducing a new temporal dimension for hyperpolarized NMR studies of cellular metabolism and enabling rapid

Photooxygenation of Heterocycles

Current Organic Chemistry, 2005
The photooxygenation of heterocycles represents a versatile and widely accepted tool for introducing oxygenated functions in a mild, simple and selective way. The review evidences the synthetic potential of the photooxygenation with particular attention to the reaction of Type II involving singlet oxygen in the first electronically excited state ( 1 Δg)

Heterocyclic NO prodrugs

Il Farmaco, 1999
An overview of the different heterocyclic NO-releasing compounds is given. Mesoionic heterocycles like sydnone imines are one example. This class is discussed on the synthesis and the mechanism of NO formation from Molsidomine and its first metabolite SIN-1.

Heterocyclic Quinodimethanes

Topics in Current Chemistry, 2017
This chapter describes a series of heterocyclic quinodimethanes mainly containing O, N, S and Si atoms in their frameworks with either closed- or open-shell electronic structures in the ground state. Their syntheses, structural characterizations and chemical and physical properties are comprehensively reviewed.

Organohypervalent heterocycles

Chemical Society Reviews
This review summarizes structural and synthetic aspects of heterocyclic molecules incorporating an atom of a hypervalent main-group element. The higher thermal stability of hypervalent heterocycles, as compared to their acyclic analogs, adds special feature to their chemistry.
