Post‐translational modifications of histones: Mechanisms, biological functions, and therapeutic targets

open access: yesMedComm, 2023
Histones are DNA‐binding basic proteins found in chromosomes. After the histone translation, its amino tail undergoes various modifications, such as methylation, acetylation, phosphorylation, ubiquitination, malonylation, propionylation, butyrylation ...

Spatial Control of Light‐Responsive Proteins and Optogenetics Within Hydrogels via Volumetric Bioprinting

open access: yesAdvanced Materials, EarlyView.
Spatiotemporal control over cell behavior within bioprinted constructs remains a key challenge in biofabrication. Volumetric bioprinting using multi‐wavelength light sources permits to activate light‐sensitive intracellular proteins as well as optogenetic gene and protein expression circuits in bioprinted cells, in addition to enabling fast printing of

Chemical Screening Identifies Danofloxacin as a Self‐Renewal Agonist of Embryonic Stem Cells through Alleviation of HDAC1‐Mediated Deacetylation of Tert and Prdm10

open access: yesAdvanced Science, EarlyView.
This study identified Danofloxacin as a novel supporter of the maintenance of mouse embryonic stem cell (ESC) pluripotency. It functions partially by inhibiting HDAC1, which increases specific histone acetylation marks and activates key genes like Tert and Prdm10.

The Growth, Pathogenesis, and Secondary Metabolism of Fusarium verticillioides Are Epigenetically Modulated by Putative Heterochromatin Protein 1 (FvHP1)

open access: yesJournal of Fungi
Fusarium verticillioides is a globally prevalent phytopathogenic fungus responsible for multiple diseases in maize and a major producer of the mycotoxin fumonisin B1 (FB1), a highly toxic fungal secondary metabolite (FSM).

Gm26550 Modulates Learning and Memory by Increasing IGF1 mRNA Expression and Stability in Nrf2−/− Mice

open access: yesAdvanced Science, EarlyView.
The present study shows that Nrf2 directly binds to the Gm26550 promoter, thereby activating Gm26550 transcription and increasing its expression. Mechanistically, Gm26550 promotes IGF1 expression by functionally antagonizing miR‐26a‐5p‐mediated repression and sequestering the RBP KHSRP, thereby enhancing hippocampal neuronal synaptic plasticity and ...

Modulating Cognition-Linked Histone Acetyltransferases (HATs) as a Therapeutic Strategy for Neurodegenerative Diseases: Recent Advances and Future Trends

open access: yesCells
Recent investigations into the neuroepigenome of the brain are providing unparalleled understanding into the impact of post-translational modifications (PTMs) of histones in regulating dynamic gene expression patterns required for adult brain cognitive ...

Genetic Ablation and Multi‐Omics Profiling Reveal CEP55 as a Key Driver of Tumorigenesis in Diverse Cancer Models

open access: yesAdvanced Science, EarlyView.
Genetic ablation of Cep55 in Pten‐deficient mouse models delays tumorigenesis. Integrated multi‐omics analyses (proteomics, phosphoproteomics, and spatial transcriptomics) reveal that CEP55 regulates oncogenic signaling (RAS/ERK, PI3K/AKT), integrin/FAK‐mediated adhesion, extracellular matrix (ECM) remodeling, and endocytosis.

Chromatin Packing Domain Engineering Through the Manipulation of Nuclear Cationic States

open access: yesAdvanced Science, EarlyView.
Manipulation of intranuclear divalent cation concentrations rapidly and reversibly remodels chromatin packing domains in living cells. Magnesium depletion disrupts domain compaction, heterochromatin organization, and transcriptional regulation, whereas magnesium loading promotes more compact and stable domains.

A Personalized Haplotype‐Resolved Near‐Gapless Genome Framework for Somatic Variant Discovery in Hepatocellular Carcinoma

open access: yesAdvanced Science, EarlyView.
A patient‐specific, haplotype‐resolved genome framework improves detection and interpretation of somatic variants in hepatocellular carcinoma. Using multi‐platform sequencing, the personalized assembly resolves complex regions including centromeres and MHC/HLA loci, enhances structural variant discovery, and links regulatory alterations to allele ...

Another twist in the histone memory code [PDF]

open access: yesCell Research, 2014
Transcription is a highly regulated process and several studies have examined the role of transcription factors and various epigenetic regulators in memory formation. In a recent paper in Nature, Zokvic and colleagues show an important role for a novel regulator of chromatin structure (the incorporation of histone variants) in memory formation.
