Time‐Controlled Refrigerated Stem Cell Therapy Mitigates Scleroderma Fibrosis via Modulation of Mitochondrial Autophagy and Gut Metabolism

This study established an RT‐MSCs‐based therapeutic approach for scleroderma in mice. RT‐MSCs attenuated fibrosis by regulating mitochondrial autophagy and restored gut microbiota homeostasis. Metabolomic analyses confirmed recovery of key metabolites, and RT‐MSCs demonstrated favorable lesion targeting and safety profiles.

JmjC histone demethylases act as chromatin oxygen sensors

Oxygen sensing is important in physiology but also in disease. We find that hypoxia (oxygen deficiency) triggers rapid and hypoxia-inducible factor (HIF)-independent histone methylation changes which are reversible upon reoxygenation.

ECM‐Stiffness Mediated Persistent Fibroblast Activation Requires Integrin and Formin Dependent Chromatin Remodeling

Prolonged exposure to stiff extracellular matrix drives cancer‐associated fibroblasts into a persistently activated myofibroblast state. Two parallel pathways are identified: β1 integrin activation smoothens the nuclear lamina to reduce lamin–chromatin contacts, while the formin mDia2 regulates nuclear actin to alter chromatin organization.
