HIV-1 detection in the olfactory mucosa of HIV-1-infected participants

open access: yesAIDS, 2019
HIV infection chronically affects the central nervous system (CNS). Olfactory mucosa is a unique site in the respiratory tract that is directly connected to the CNS; thus we wanted to evaluate olfactory mucosa as a surrogate of CNS sampling. We conducted a preliminary study examining HIV populations and susceptible cells in the olfactory mucosa ...

Single genome sequencing of near full-length HIV-1 RNA using a limiting dilution approach [PDF]

open access: yes, 2019
Sequencing very long stretches of the HIV-1 genome can advance studies on virus evolution and in vivo recombination but remains technically challenging.

Human Schlafen 14 Cleavage of Short Double‐Stranded RNAs Underpins its Antiviral Activity

open access: yesAdvanced Science, EarlyView.
SLFN14 is associated with human diseases. SLFN14 is found to cleave RNAs containing short duplexes. The cryo‐EM structures of SLFN14 and SLFN14‐hairpin RNA complex reveal that SLFN14 assembles into a ring‐like dimer; two RNase domains form an RNA‐binding groove accommodating a hairpin RNA.

Early Therapy in HIV-1-Infected Children: Effect on HIV-1 Dynamics and HIV-1-Specific Immune Response

open access: yesAntiviral Therapy, 2008
Background Perinatal HIV-1 infection is acquired in the milieu of a developing immune system, leading to high levels of uncontrolled viral replication. Few data have been reported that address the viral dynamics and immunological response in infants who initiated aggressive antiretroviral therapy (ART) shortly after birth.

Generating Vectors for the Lattice Structures of Tubular and Conical Viral Capsids

open access: yesComputational and Mathematical Biophysics, 2014
Retrovirus capsid is a fullerene-like lattice consisting of capsid protein hexamers and pentamers. Mathematical models for the lattice structure help understand the underlying biological mechanisms in the formation of viral capsids.

Variations in Innate Immune Cell Subtypes Correlate with Epigenetic Clocks, Inflammaging and Health Outcomes

open access: yesAdvanced Science, EarlyView.
This study uses a high‐resolution DNA methylation reference panel for 19 immune cell‐types, as well as transcriptomic, metabolomic, and large population cohort data to show that age‐related epigenetic heterogeneity within monocytes correlates with inflammaging, biological age, and all‐causemortality, and does so more strongly than existing monocyte ...

Combination therapy with anti-HIV-1 antibodies maintains viral suppression

open access: yesNature, 2018
Individuals infected with HIV-1 require lifelong antiretroviral therapy, because interruption of treatment leads to rapid rebound viraemia. Here we report on a phase 1b clinical trial in which a combination of 3BNC117 and 10-1074, two potent monoclonal ...

CLAE: A High‐Fidelity Nanopore Sequencing Strategy for Read‐Level Viral Variant Detection and Environmental RNA Virus Discovery

open access: yesAdvanced Science, EarlyView.
High‐fidelity Nanopore sequencing offers a cost‐effective, portable path to uncovering viral dark matter in complex environments—but suffers from severe read‐length bias, low throughput, and limited accuracy. This study traces these limitations to core biochemical barriers and introduces CLAE, a foundational high‐fidelity platform that enables accurate,
