The preparation of trisubstituted hydantoins features a Mitsunobu reaction for introduction of the substituent on N-1.

Five unrelated children born to epileptic women treated with hydantoin anticonvulsants were found to have a similar broad multi-system pattern of abnormalities, including craniofacial anomalies, nail and digital hypoplasia, prenatal-onset growth deficiency, and mental deficiency.

Hydantoins as antitumor agents

A series of 27 hydantoins was prepared and tested as antitumor agents. These were variously substituted in the 5 position but with special emphasis on the substituents (chloro, acetyl, chloroacetyl, and methyl) in the 1 and/or 3 positions.
