The hydroxyectoine gene cluster of the non-halophilic acidophile Acidiphilium cryptum

Acidiphilium cryptum is an acidophilic, heterotrophic α-Proteobacterium which thrives in acidic, metal-rich environments (e.g. acid mine drainage). Recently, an ectABCDask gene cluster for biosynthesis of the compatible solutes ectoine and hydroxyectoine was detected in the genome sequence of A. cryptum JF-5.

Ectoine is an osmoprotector that is synthesized by many halophilic and halotolerant bacteria in response to osmotic stress. The halotolerant methanotroph Methylomicrobium alcaliphilum 20Z was genetically modified to produce ectoine derivative hydroxyectoine by introduction of the gene encoding ectoine hydroxylase EctD from Pseudomonas stutzeri DSM5190T.

Downshock treatment of the halotolerant Brevibacterium sp. JCM 6894 was a prerequisite for proline uptake which is a function for cell survival. Hydroxyectoine served as an effective stimulator for the proline uptake and cell survival of the downshocked cells of this strain.
