Asian diets high in soy are associated with lower risk for breast cancer compared with Western diets. Moreover, higher levels of two putative carcinogenic metabolites of 17beta-estradiol, 4- and 16alpha-hydroxyestrogen, and lower amounts of anticarcinogenic metabolites, 2-hydroxyestrogens, have been associated with greater breast cancer risk.

The method is highly specific, accurate and precise within the range of 0.2-20 nmol 2-hydroxyestrone/20ml of urine.

2-hydroxyestrone: the 'good' estrogen.
The Journal of endocrinology, 1996The issue of the role of 2-hydroxyestrone (2-OHE1) in breast cancer has been the subject of considerable controversy as to whether it is carcinogenic or anticarcinogenic. The expanding data base outlined below is most consistent with the conclusion that 2-OHE1 is anticarcinogenic.

Approximately one-third to one-half of individuals harbor the colonic bacteria that are capable of metabolizing the soy isoflavone daidzein to equol. Results of prior studies suggest beneficial effects of producing equol in relation to breast cancer risk, potentially through effects on endogenous hormones.

Structure of lysine adducts with 16α-hydroxyestrone and cortisol
Journal of Steroid Biochemistry, 1986Recent studies indicate that steroids containing a vicinal hydroxyketone moiety can react with proteins both in vitro and in vivo to form covalent addition products. This reaction is non-enzymatic and occurs via the Heyns rearrangement of an initial Schiff base adduct between the steroid carbonyl and the epsilon-amino group of lysine residues.

Metabolic Clearance Rate and Uterotropic Activity of 2-Hydroxyestrone in Rats*
Endocrinology, 19812-Hydroxyestrone (2-OHE1) has much lower uterotropic potency than might be predicted from its uterine estrogen receptor affinity. 2-OHE1 displaces saturably bound [3H]estradiol from rat uterine cytosol with a competitive inhibition constant of 8.6 nM, while the dissociation constant for 17 beta-estradiol (E2) is 0.42 nM.

Species difference in the structure of urinary 2-hydroxyestrone “glucuronide”
Steroids, 1970Abstract Metabolism of radioactive estrone by female rats and hamsters leads to excretion of urinary 2-hydroxyestrone conjugated with “glucuronic acid” exclusively at the 2 position. Simular experiments with the human are reported to give 2-hydroxyestrone conjugated at the 3 position (Yoshizawa and Fishman, J. Clin. Endocrin. Metab. 29 , 1123 (1969).
