Growth-stimulated synchronized cells exhibit a rapid increase in 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMG-CoA reductase, EC 1.1.1.88) activity prior to the onset of DNA synthesis. Under normal culture conditions, HMG-CoA reductase activity exhibits wide variations among experiments.

Abstract The activity of rat liver microsomal hydroxymethylglutaryl CoA reductase (HMG CoA reductase, E.C. 1.1.1.34) is severely diminished when microsomes are preincubated with both ATP and Mg 2+ before the enzyme is assayed. Sensitivity to MgATP is progressively lost when microsomes are extracted with buffers.

The reaction catalyzed by 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase [hydroxymethylglutaryl-CoA reductase (NADPH), EC 1.1.1.34] involves reductive deacylation of HMG-CoA to mevalonate and requires two moles of NADPH (Fig. 1).
