Targeting Lactate and Lactylation in Cancer Metabolism and Immunotherapy

open access: yesAdvanced Science, EarlyView.
Lactate, once deemed a metabolic waste, emerges as a central regulator of cancer progression. This review elucidates how lactate and its epigenetic derivative, protein lactylation, orchestrate tumor metabolism, immune suppression, and therapeutic resistance.

Implementing the 2013 WHO diagnostic criteria for gestational diabetes mellitus in a Rural Nigerian Population

open access: yesThe Pan African Medical Journal, 2020
INTRODUCTION: The World Health Organization (WHO) reviewed the threshold values required for the diagnosis of Gestational Diabetes Mellitus (GDM) in 2013 and the implementation of the new diagnostic criteria have been associated with increase in the ...

SOCS3 drives proteasomal degradation of indoleamine 2,3-dioxygenase (IDO) and antagonizes IDO-dependent tolerogenesis [PDF]

open access: yesProceedings of the National Academy of Sciences, 2008
Despite their common ability to activate intracellular signaling through CD80/CD86 molecules, cytotoxic T lymphocyte antigen 4 (CTLA-4)-Ig and CD28-Ig bias the downstream response in opposite directions, the latter promoting immunity, and CTLA-4-Ig tolerance, in dendritic cells (DCs) with opposite but flexible programs of antigen ...

Breast Cancer Chemo-immunotherapy through Liposomal Delivery of an Immunogenic Cell Death Stimulus Plus Interference in the IDO-1 Pathway

open access: yesACS Nano, 2018
Immunotherapy provides the best approach to reduce the high mortality of metastatic breast cancer (BC). We demonstrate a chemo-immunotherapy approach, which utilizes a liposomal carrier to simultaneously trigger immunogenic cell death (ICD) as well as ...

Trial watch: IDO inhibitors in cancer therapy

open access: yesOncoimmunology, 2020
Indoleamine 2,3-dioxygenase 1 (IDO1) catalyzes the first, rate-limiting step of the so-called “kynurenine pathway”, which converts the essential amino acid L-tryptophan (Trp) into the immunosuppressive metabolite L-kynurenine (Kyn).

Non‐Targeting shRNA‐Encoded Plasmid DNA Enhances Protective Immunity Through RIDD‐RIG‐I Signaling Pathway in the Zika Virus Animal Model

open access: yesAdvanced Science, EarlyView.
We present a novel DNA vaccine platform featuring intrinsic, non‐targeting dsRNA that significantly enhances immune responses by activating the regulated IRE1‐dependent decay‐RIG‐I signaling pathway. This research elucidates a previously uncharacterized mechanism for dsRNA‐mediated innate immune activation.

Self-medication among health workers in a tertiary institution in South-West Nigeria

open access: yesThe Pan African Medical Journal, 2016
INTRODUCTION: inappropriate self-medication results in wastage of resources, resistance to pathogen and generally entails serious health hazard. This study was undertaken to determine the knowledge, practice and reasons for practice of self-medication ...

Blockade of IDO-kynurenine-AhR metabolic circuitry abrogates IFN-γ-induced immunologic dormancy of tumor-repopulating cells

open access: yesNature Communications, 2017
Interactions with the immune system may lead tumorigenic cells into dormancy. However, the underlying molecular mechanism is poorly understood. Using a 3D fibrin gel model, we show that IFN-γ induces tumour-repopulating cells (TRCs) to enter dormancy ...

Biomimetic Iridium‐Based Photothermal Nanozyme to Trigger Ferroptosis and Pyroptosis and Activate the cGAS‐STING Pathway for Improved Tumor Immunotherapy

open access: yesAdvanced Science, EarlyView.
This study develops an iridium‐based nanozyme (IIN@M) with superior enzyme‐like activity and photothermal performance. It disrupts intracellular redox balance by generating reactive oxygen species and depleting glutathione, which synergistically induces ferroptosis, pyroptosis, and immunogenic cell death.
