The therapeutic effects of labetalol in chronic stress- and immuno-agonist-induced cough hypersensitivity and lung inflammation involve reducing interferon-γ-producing T lymphocytes

open access: yesERJ Open Research
Background Psychological stress correlates with chronic cough, which is often triggered by respiratory viruses. Chronic stress promotes sympathetic nerves to release norepinephrine and neuropeptide Y.

IFN-γ-Induced intestinal epithelial cell-type-specific programmed cell death: PANoptosis and its modulation in Crohn’s disease

open access: yesFrontiers in Immunology
BackgroundCrohn’s disease (CD) is a chronic inflammatory bowel disease (IBD) and is considered a Th1-mediated disease, supported by the over-expression of interferon-gamma (IFN-γ) in the intestinal lamina propria.

Single‐Cell Dissection of Therapy‐Induced Remodeling Uncovers a Fibroblast‐Driven Immunosuppressive Niche and Targetable Vulnerabilities in Lethal Prostate Cancer

open access: yesAdvanced Science, EarlyView.
Single‐cell longitudinal profiling reveals that androgen‐deprivation therapy induces a DPT+ fibroblast‐complement axis that suppresses macrophage inflammation and drives CD8+ T cell exhaustion in prostate cancer. Concurrently, resistant epithelial subpopulations persist and engage TSPAN1‐ and NRXN1‐mediated programs promoting CRPC and neuroendocrine ...

RBM10 Deficiency Promotes Anti‐PD‐1 Resistance in LUAD via STING Alternative Splicing‐Driven CCL7 Signaling and Macrophage Polarization

open access: yesAdvanced Science, EarlyView.
RBM10 deficiency promotes anti‐PD‐1 resistance in lung adenocarcinoma by altering STING alternative splicing, which enhances CCL7 secretion and CCR2‐dependent M2 macrophage polarization. A positive feedback loop via mitochondrial transfer sustains this immunosuppression.

Aptamer‐Engineered Liposomal Platform Enables in Situ cDC1 Vaccination to Potentiate Immunotherapy in Prostate Cancer

open access: yesAdvanced Science, EarlyView.
Prostate cancer is immunologically ‘cold’, with scarce, dysfunctional type 1 conventional dendritic cells (cDC1s) that limit T cell priming. We introduce an aptamer‐targeted liposomedelivering FMS‐like tyrosine kinase 3 ligand (Flt3L) and chlorin e6 (Ce6). Ultrasound induces antigen release and cDC1s recruitment, creating an in situ cDC1 vaccine.

Engineering Approaches to Modify Immunomodulatory Functions of Mesenchymal Stromal Cells (MSCs): Tissue Regeneration and Clinical Application

open access: yesAdvanced Science, EarlyView.
Mesenchymal stromal cells (MSCs) show promise for treating immune‐related disorders through immunomodulation and tissue regeneration. This review gives a brief overview of current clinical approval of MSC therapies. It also discussed how bioengineering, including genetic modification, biomaterial delivery, extracellular vesicles, and iPSC‐derived MSCs,

Cigarette Smoke‐Induced Alveolar Macrophage Senescence via GEM/SIRT3‐Mediated Mitochondrial Dysfunction

open access: yesAdvanced Science, EarlyView.
Cigarette smoke accelerates the aging of immune cells in the lung. By combining human single‐cell sequencing, cell culture, and mouse models, the authors show that the protein GEM drives mitochondrial damage and senescence in alveolar macrophages by suppressing SIRT3.

NKG2D modulates aggravation of liver inflammation by activating NK cells in HBV infection

open access: yesScientific Reports, 2017
Hepatitis B virus (HBV) infection is thought to be an immune-mediated liver disease. The mechanisms underlying natural killer (NK) cell group 2D receptor (NKG2D) that activates NK cells and participates in anti-HBV immunity and immunopathology has not ...

TME/NIR Dual‐Responsive Zinc‐Based Targeted Nanoagonist for Multimodal Amplification of STING‐Mediated Cancer Immunotherapy

open access: yesAdvanced Science, EarlyView.
A multifunctional nanoagonist (cDZ@IP) enables nano‐metabolite–driven multimodal activation of the STING pathway and enhanced immune recognition, achieving potent antitumor immunity and suppressing tumor growth and metastasis. This strategy highlights the rational design of therapeutic metabolites and establishes a new paradigm bridging nanomedicine ...

Radiotherapy Enhances the Oncolytic Efficacy of the Novel Oncolytic Herpesvirus VG161 and Amplifies Its Antitumor Immunity in Breast Cancer

open access: yesAdvanced Science, EarlyView.
RT promotes VG161 replication in BC and its immunostimulatory transgenes expression, which is mediated by the upregulation of GADD34 and HVEM caused by RT. The combination therapy with VG161 and RT increases the abundances of tumor‐infiltrating lymphocytes and elicits potent systemic antitumor immunity, thereby effectively inhibiting local tumors and ...
