ABSTRACT Objective Amyotrophic lateral sclerosis (ALS) is characterized by progressive motor neuron degeneration. Glycolytic dysregulation is implicated in disease progression, yet the underlying mechanisms remain unclear. This study investigates how Aldolase A (ALDOA) drives ALS progression through glycolysis‐mediated motor neuron pyroptosis.

GCase activity in mice receiving diltiazem by intraperitoneal injection:
The 9H mice were given diltiazem by intraperitoneal injections at 50, 100 or 200 mg/kg/d for 7 days. GCase activity increased by ∼1.2 fold in the brain of D409H mice at each of the three doses, i.e., the response was not dose dependent.

A biomimetic hydrogel derived from neonatal spinal cord ECM (DNSCM) delivers LAMB2 protein and ISP peptide to remodel the inhibitory injury microenvironment. The system sustains drug release, upregulates pro‐regenerative LAMB2, downregulates inhibitory CSPG, and promotes axonal regeneration.
