Configurational Differences and Binding Mechanisms of Interleukin-1 Receptor-Associated Kinase 1
2020 IEEE 20th International Conference on Bioinformatics and Bioengineering (BIBE), 2020Interleukin-l receptor-associated kinase 1 (IRAKI) is crucial for downstream regulation of the toll-like receptor signaling pathway and is involved in innate immune system, inflammatory diseases, and cancers. There is an urgent need for developing inhibitors that target IRAKI.

Pathophysiology of interleukin-1 receptor-associated kinase-M: implications in refractory state
Current Opinion in Infectious Diseases, 2006The pseudo-kinase interleukin-1 receptor-associated kinase-M has emerged as a critical molecule in the down-regulation of inflammatory responses. Dysregulation of the toll-like receptor-interleukin-1 receptor-associated kinase system, and in particular interleukin-1 receptor-associated kinase-M up-regulation, are associated with a number of pathologies.

Discovery and initial SAR of inhibitors of interleukin-1 receptor-associated kinase-4
Bioorganic & Medicinal Chemistry Letters, 2006High-throughput screening of a small-molecule compound library resulted in the identification of a novel series of N-acyl 2-aminobenzimidazoles that are potent inhibitors of interleukin-1 receptor-associated kinase-4.

Small Molecule Inhibition of Interleukin-1 Receptor-Associated Kinase 4 (IRAK4)
2017In recent years, interleukin-1 receptor-associated kinase 4, IRAK4, has become an attractive target for many medicinal chemistry programmes. Target inhibition is of potential therapeutic value in areas including autoimmune disorders, cancer, inflammatory diseases, and possibly neurodegenerative diseases.

Differential regulation of interleukin-1 receptor associated kinase 1 (IRAK1) splice variants
Molecular Immunology, 2007IRAK family proteins play critical roles in regulating innate immunity. There are three differentially spliced variants of IRAK1, namely 1, 1b, and 1c. We and others have previously identified that the full length IRAK1 underwent covalent modifications such as phosphorylation and ubiquitination upon lipopolysaccharide challenge.

AbstractEndometrial carcinoma (EC) is one of the most common malignancies in the world. Previous studies have investigated the altered expression of interleukin‐1 receptor‐associated kinase 1 (IRAK1) in various cancers. We aimed at exploring the biological function and the underlying molecular mechanism of IRAK1 in EC.

The interleukin-1 receptor associated kinases (IRAKs) are critically involved in the IL-1R/Toll-like receptor (TLR)-mediated signal transduction processes and therefore regulate cellular innate immune responses. Four IRAK members have been identified in the human genome (IRAK-1, 2, M, and 4), which seem to play distinct roles.
