Electrical pulse stimulation reflecting the episodic nature of real‐life exercise modulates metabolic and secretory profile of primary human myotubes

Here, we introduced an intermittent electrical stimulation protocol mimicking the episodic nature of real‐life exercise in vitro by alternating low‐ and high‐frequency stimulation. In comparison with widely used continuous stimulation, it enhanced the rate of glucose and fatty acid oxidation, but not the myokine release.

Tryptophan metabolites were analyzed across twelve organs, the central nervous system, and serum in male and female mice at three life stages. We found tissue‐, sex‐, and age‐specific differences, including increased indole‐3‐pyruvate and kynurenine in aging males.

Indoleamine 2,3‐dioxygenase 1 inhibition reverses cancer‐associated fibroblast‐mediated immunosuppression in high‐grade serous ovarian cancer

CAF‐mediated immunosuppression in ovarian cancer is driven by IDO1, reducing T‐cell function. Inhibiting IDO1 restores T‐cell proliferation and cytotoxicity, increases cancer cell apoptosis, and may help overcome CAF‐induced immune suppression in high‐grade serous ovarian cancer. Targeting IDO1 may improve antitumor immunity.

Interleukin-6 is a multifunctional cytokine produced by variety of cells including tumor cells. It is produced by monocytes, macrophages, B-cells and T-cells. Interleukin-6 stimulates platelet production through thrombopoietin. Interleukin-6 in surplus amount is also produced by epithelial ovarian cancer cells.
