Elevated SPARC Disrupts the Intestinal Barrier Integrity in Crohn's Disease by Interacting with OTUD4 and Activating the MYD88/NF‐κB Pathway

open access: yesAdvanced Science, Volume 12, Issue 11, March 20, 2025.
This study reveals that SPARC activates the MYD88/NF‐κB/MLCK/MLC2 pathway by interacting directly with OTUD4, leading to intestinal barrier damage. In addition, the METTL3‐YTHDF1 axis promotes SPARC expression. Therefore, targeting SPARC or the OTUD4/MYD88/NF‐κB/MLCK/MLC2 axis may provide new insights into the molecular basis of CD and may serve as an ...

Chromosome Missegregation Triggers Tumor Cell Pyroptosis and Enhances Anti‐Tumor Immunotherapy in Colorectal Cancer

open access: yesAdvanced Science, Volume 12, Issue 12, March 27, 2025.
Immune system disorders and chromosome missegregation are two of the major hallmarks of tumor. The relationship between tumor immune and chromosome missegregation is still unclear. This study reveals that YY2 high expression‐mediated chromosome missegregation/micronuclei formation triggers pyroptosis and CTL activation by activating AIM2/caspase‐1 ...

Intestinal Gastrin/CCKBR Axis Protects against Type 2 Diabetes by Reducing Intestinal Glucose Absorption through the PI3K/Akt/eIF4B Signaling Pathway

open access: yesAdvanced Science, Volume 12, Issue 13, April 3, 2025.
In intestinal epithelial cells, Gastrin/CCKBR stimulation prevents high glucose‐stimulated SGLT1 (SLC5A1) and GLUT2 (SLCA2) expressions, via the PI3K/AKT/eIF4B signaling pathway, with facilitatory effects on the secretion of incretins, which could be therapeutic targets for clinical treatment in T2D.

Muscle‐Derived Small Extracellular Vesicles Mediate Exercise‐Induced Cognitive Protection in Chronic Cerebral Hypoperfusion

open access: yesAdvanced Science, EarlyView.
sEVs have a critical role in orchestrating interorgan crosstalk and mediating exercise‐induced therapeutic effects. Lin et al. demonstrates that sEVs miR‐17/20a‐5p mediates the muscle‐brain crosstalk and emphasizes the central role of mTOR signaling in executing molecular programs that can protect brain health in response to exercise. Abstract Physical

Rational Design of Inner Ear Drug Delivery Systems

open access: yesAdvanced Science, EarlyView.
Hearing loss is a common disease affecting many people, and inner ear lesions are one of the most important causes. This review focuses on the treatment of inner ear hearing loss by drug delivery systems. It includes the current methods and technologies developed, and it predicts possible directions.
