Macrophage polarization by apoptotic cancer cells - a RNAi high-throughput screen and validation of interleukin 10 regulation [PDF]

open access: yes, 2012
Tumor-associated macrophages (TAM) are a major supportive component within neoplasms and by their plasticity promote all phases of tumor development. Mechanisms of macrophage (M Phi) attraction and differentiation to a tumor-promoting phenotype, defined ...

Copper‐Driven Epithelial Barrier Disruption: A Novel Mechanism of COPD Acute Exacerbations Mediated by the TNF‐α/ATP7A Axis

open access: yesAdvanced Science, EarlyView.
This study reveals a copper‐driven mechanism of COPD acute exacerbations mediated by the TNF‐α/ATP7A axis. TNF‐α suppresses the copper transporter ATP7A via competitive NF‐κB/CREB1/CBP signaling, resulting in intracellular copper accumulation, mitochondrial oxidative stress, and epithelial barrier disruption.

Innate Immunocompetent hiPSC‐Derived Neurospheroids Capture Early CNS Responses to rAAV

open access: yesAdvanced Science, EarlyView.
Knowledge of human CNS immune responses to AAV‐based gene therapies remains limited due to the lack of immune‐competent human models. Here, a hiPSC‐derived 3D neuroimmune platform integrating neurospheroids and microglia is established using stirred‐tank bioreactors.

Characterisation of keratinocyte-derived epidermal signals in the initiation of contact hypersensitivity to chemicals [PDF]

open access: yes, 2015
Contact hypersensitivity reactions result from an adaptive immune response following topical exposure to chemical allergen. The initiation of this response requires the production of an inflammatory micro-environment, however the contribution of ...

Synthetic Toll‐Like Receptors for Control of Innate Immunity With Far‐Red Light

open access: yesAdvanced Science, EarlyView.
Synthetic Toll‐like receptors (TLRs) are engineered by replacing their natural sensing regions with a far‐red‐light‐responsive module from a bacterial phytochrome. These synthetic receptors enable light‐controlled regulation of immune signaling in mammalian cells.

TRAIP Mediates Alcohol‐Induced Liver Injury through Regulating β‐catenin Ubiquitin Degradation via Direct Interaction

open access: yesAdvanced Science, EarlyView.
This study reveals that NF‐κB1‐driven TRAIP upregulation in ALD correlates with disease severity. Mechanistically, TRAIP directly binds β‐catenin via its CC domain and promotes its K48‐linked ubiquitination and degradation, which is independent of the GSK3β/β‐TrCP pathway.

Disrupted TFEB/GDNF–cAMP/ATP Coupling Underlies Astrocytic Dysfunction and Depression in LRRK2 G2019S Parkinson's Mice

open access: yesAdvanced Science, EarlyView.
ABSTRACT The LRRK2 G2019S mutation, a Parkinson's disease–linked variant, has been associated with depression‐like phenotypes, but mechanisms remain unclear. We chart age‐dependent behavioral changes and astrocyte reactivity in G2019S mice and define a pathway connecting LRRK2 to TFEB/GDNF signaling, cellular energetics, and inflammation.

Altered Expression of Cell Cycle Regulators and Factors Released by Aged Cells in Skeletal Muscle of Patients with Bone Fragility: A Pilot Study on the Potential Role of SIRT1 in Muscle Atrophy

open access: yesBiomedicines
Background/Objectives: Cellular aging represents a crucial element in the progression of musculoskeletal diseases, contributing to muscle atrophy, functional decline, and alterations in bone turnover, which promote fragility fractures. However, knowledge

The impact of interleukin-6 receptor inhibitors on risk of diabetes mellitus in patients with giant cell arteritis: a cohort study

open access: yesRheumatology International
Abstract To investigate the one-year incidence of diabetes mellitus (DM) among patients with giant cell arteritis (GCA) treated with glucocorticoids and interleukin-6 receptor inhibitor (IL-6Ri) compared to patients treated with glucocorticoid monotherapy.
