In Situ Programmable Modulation of Hydrogel Stiffness for Stage‐Adaptive Bone Regeneration

open access: yesAdvanced Science, EarlyView.
Near‐infrared (NIR) irradiation triggers Ca2+ release from thermoresponsive nanocarriers, enabling in situ stiffening of an adaptive hydrogel from a soft to a stiff state, thereby sequentially supporting mesenchymal stromal cell stemness and osteogenic differentiation for stage‐adaptive bone regeneration.

ECM stiffness regulates calcium influx into mitochondria via tubulin and VDAC1 activity

open access: yesAnimal Cells and Systems
Calcium ions (Ca2+) play pivotal roles in regulating numerous cellular functions, including metabolism and growth, in normal and cancerous cells. Consequently, Ca2+ signaling is a vital determinant of cell fate and influences both cell survival and death.

Interactions of intracellular calcium and immune response in earthworms [PDF]

open access: yesInvertebrate Survival Journal, 2011
Intracellular calcium level has a definite role in innate and adaptive immune signaling but its evolutionary aspects are not entirely clear yet. Very few information are accessible about calcium contents of invertebrate immunocytes, especially of ...

Ion‐Driven Regulation of Ca2+ Flow by Ti/Zn Composite Activates ERK/MAPK Signaling and Maintains Mitochondrial Homeostasis to Promote Macrophage‐Mediated Bone Regeneration

open access: yesAdvanced Science, EarlyView.
A 3D‐printed interpenetrating Ti/Zn composite enables sustained Zn2+ release to regulate intracellular Ca2+ redistribution, preserve mitochondrial homeostasis, activate ERK/MAPK signaling, and promote M2 macrophage polarization, thereby enhancing osteoimmune‐mediated bone regeneration and peri‐implant osseointegration.

A Novel Pak1 Activator Ameliorates ER Stress for HFpEF Therapy

open access: yesAdvanced Science, EarlyView.
Chronic metabolic stress is a major contributor to HFpEF progression. Under prolonged metabolic stress, Pak1 activity becomes impaired, contributing to disrupted ER proteostasis, cardiomyocyte apoptosis, fibrosis, and diastolic dysfunction. Mechanistically, Pak1 overexpression activates the ERK1/2–MNK1–eIF4E signaling axis, promotes translational ...

ATL2 Recruits TRAK1 to Promote Mitochondrial Transport at ER–Mitochondria Contact Sites

open access: yesAdvanced Science, EarlyView.
ABSTRACT Mitochondrial transport and distribution are crucial for cellular homeostasis, yet whether and how they are regulated by endoplasmic reticulum (ER)–mitochondria contact sites remains unclear. Here, we demonstrate that the ER protein atlastin‐2 (ATL2) orchestrates mitochondrial transport and distribution by promoting assembly of the transport ...

Calcium oscillations and mitochondrial enzymes in stem cells

open access: yesRegenerative Therapy
Calcium oscillations are rhythmic fluctuations of the intracellular concentration of calcium ions (Ca2+). As Ca2+ evokes various cellular processes, its intracellular concentration is tightly regulated.

A Rare De Novo Missense Mutation in IFT122 Confers a Genetic Susceptibility Factor of Idiopathic Pediatric Uveitis Via Trio‐based Whole‐Exome Sequencing

open access: yesAdvanced Science, EarlyView.
A rare de novo IFT122‐A773E variant is identified in idiopathic pediatric uveitis and shown to exacerbate retinal inflammation and barrier dysfunction. Mechanistically, the variant enhances IFT43 interaction, elevates calcium signaling, and activates the MEK/ERK/FRA1 axis, revealing a previously unrecognized cilia‐associated pathway that may increase ...

Transcription Factor HOXC11 Drives Colorectal Cancer Progression and Metastasis via CAMK2A‐Dependent CXCL5 Upregulation

open access: yesAdvanced Science, EarlyView.
HOXC11 drives colorectal cancer progression by transcriptionally activating CAMK2A, which triggers NF‐κB–dependent CXCL5 upregulation. CXCL5–CXCR2 signaling further reinforces HOXC11 expression through the ERK1/2–SP1 axis, forming a prometastatic positive feedback loop that is effectively disrupted by combined CAMK2A and CXCR2 inhibition.
