Immobilization of inulinase for sucrose hydrolysis

A commercial inulinase preparation immobilized on various supports was used for sucrose hydrolysis. Entrapment and encapsulation in Ca-alginate and entrapment in an alginate–silicate sol–gel matrix were evaluated. Best results were obtained with Ca-alginate beads.

Inulinase ofDebaryomyces cantarellii

Debaryomyces cantarellii excretes into a buffered medium an inulinase of beta-fructofuranosidase type, its synthesis being induced by inulin. The enzyme has a pH optimum at 4 and its optimum temperature is 50 degrees C. Its Km for inulin is 15 mM.

[Screening strains producing inulinase and cloning of inulinase gene].

A wild-type strain AF10 producing inulinase was screened from soil samples, the strain is identified to be a A. niger. A endoinulinase gene inuA1 from A. niger AF10 was wequenced and analyzed, after PCR amplification. The results shows inuA1 is 1551 bp in length without any intron and contains 4 potential N-glycosylation sites.

Biosynthesis of Inulinases by Bacillus Bacteria

Biosynthesis of extracellular inulinase by bacteria Bacillus polymyxa 29, B. polymyxa 722, and B. subtilis 68 was studied. The optimal parameters for the producer growth were as follows: pH 7.0, 33-35 degrees C, the growth duration within 72 h. The presence of mineral reduced or of organic nitrogen was necessary for the enzyme biosynthesis.

Inulinase activity ofDebaromyces cantarellii

Debaromyces cantarellii Capriotti contains an inulinase activity which is inducible by growth on inulin but not on other beta-fructosides. The induction is inhibited by glucose and fructose. The system is situated in the cell wall and can be best extracted with a 20 mM phosphate buffer at pH 8.5.
