Comparison of anther and isolated microspore cultures in barley. Effects of culture density and regeneration medium

open access: yesEuphytica, 2000
A comparison of anther and microspore culture efficiency for green doubled haploid plant production was undertaken using 17 F1 crosses with potential agronomic performance. Green doubled haploid plants were produced from all F1 crosses by anther and microspore cultures, although there was a great variation among crosses.

Isolated microspore culture techniques and recent progress for haploid and doubled haploid plant production

open access: yesPlant Cell, Tissue and Organ Culture, 2010
An isolated microspore culture provides an excellent system for the study of microspore induction and embryogenesis, provides a platform for an ever-increasing array of molecular studies, and can produce doubled haploid (DH) plants, which are used to ...

Cytological and ultrastructural changes induced in anther and isolated-microspore cultures in barley: Fe deposits in isolated-microspore cultures

Journal of Structural Biology, 2005
To gain further insight into the role played by sporophytic anther tissues in the early stages of the androgenic process, we have compared the cytology and ultrastructure of barley embryogenic pollen grains obtained by anther culture with those obtained by isolated-microspore culture.

Barley Isolated Microspore Culture

2021
The production of doubled haploids (DHs) has proved to be a highly valuable tool to obtain new cultivars. Among the cereals, barley (Hordeum vulgare L.) is the most successful species in large-scale haploid production. Techniques employed for this purpose are based on either the gynogenetic or the androgenetic pathway.

Isolated wheat microspore culture

Plant Cell, Tissue and Organ Culture, 1995
The use of doubled haploid plants in a wheat breeding program requires an efficient haploid production system. While the techniques for producing doubled haploids from anther culture are well established, those for isolated microspores are complicated and inefficient.

Isolated Microspore Culture in Brassica napus

2020
Isolated microspore culture is the most efficient technique among those used to induce microspore embryogenesis. In the particular case of Brassica napus, it is also the most widely used and optimized. In this chapter, we describe a protocol for microspore culture in B.

Triticale Isolated Microspore Culture for Doubled Haploid Production

2021
Here, we describe a method of triticale isolated microspore culture for production of doubled haploid plants via androgenesis. We use this method routinely because it is highly efficient and works well on different triticale genotypes. To force microspores into becoming embryogenic, we apply a 21-day cold pretreatment.

Culture of Isolated Microspores of Saccharum spontaneum)

Zeitschrift für Pflanzenphysiologie, 1984
Summary Microspore culture of Saccharum spontaneum L. was investigated as a means to produce haploid plants. Isolated and cultured microspores initiated sporophytic development which in 4 to 6 weeks resulted in procalli and calli. The percentage of microspores producing multicellular procalli was increased by removing the nonviable microspores ...
