An improved in vitro technique for isolated microspore culture of barley
Euphytica, 2001A detailed procedure for isolated microspore culture of barley is presented along with examples of response across genotypes. Over 30 genotypes, including winter and spring growth habit and 2-row and 6-row genotypes, have shown an essentially genotype independent response, averaging about 10,000 embryos per 5 cm petri culture plate.

Isolated microspore culture of Canadian 6× triticale cultivars
Plant Cell, Tissue and Organ Culture, 2005Isolated microspore culture was conducted on nine Canadian triticale cultivars (X Triticosecale Wittmack) using two induction media developed for wheat, with or without 100 g l−1 Ficoll. Significant interactions were observed for the number of embryos and calluses induced, green and albino plantlets regenerated and fertility of green plants. Ficoll was

Brassica rapa L. ssp. chinensis Isolated Microspore Culture Protocol
2021Culture of isolated microspores is a widely used method to obtain haploid and doubled haploid plants for many crop species. This protocol describes the steps necessary to obtain a large number of microspore derived embryos for pakchoi (Brassica rapa L. ssp. chinensis) and zicaitai (Brassica rapa L. ssp. сhinensis Hanelt var. purpuraria Kitam).

Regeneration of fertile green plants from oat isolated microspore culture
Plant Cell Reports, 2009Regeneration of fertile green plants from isolated oat microspores is reported for the first time. Factors critical for microspore growth and regeneration include cold pre-treatment, pH of culture medium and the use of conditioned culture medium. It was found that cold pre-treatment at 4 degrees C in the dark for a minimum of 6 weeks was necessary to ...

Optimization of maize microspore isolation and culture conditions for reliable plant regeneration
Plant Cell Reports, 1991The effects of different factors were investigated in the process of isolated microspore culture of Zea mays L., Using donor plants grown in standard conditions and an efficient isolation technology, homogeneous populations of viable microspores at specific developmental stages were obtained and tested in culture.

Obtaining carrot (Daucus carota L.) plants in isolated microspore cultures
Journal of Applied Genetics, 2010Microspores were cultured on the modified B5 liquid medium containing 2.4D (0.1 mg L(-1)), NAA (0.1 mg L(-1)), L-glutamine (500 mg L(-1), L-serine (100 mg L(-1)), and sucrose (100 g L(-1)). The developmental stages of microspores and divisions were observed. Initially, the formation of binuclear and multicellular structures was noticed.

Barley isolated microspore culture (IMC) method
2003The protocol described here is for the culture of immature barley microspores, which are removed from the anther prior to culture. It is referred to as isolated microspore culture (IMC). Critical factors for success of the technique include: growing conditions for the donor plants; specific developmental stage at which microspores are collected ...

Isolation and culture of apple microspores: Some preliminary results
1994The establishment of true breeding lines is considered a critical step for cultivated variety development. Particularly in heterozygotic allogamous plants, such as apple (Malus pumila Mill), the establishment of homozygous lines offers new prospects for breeding.

Isolated microspore culture system for Brassica parachinensis
2014Brassica parachinensis is one of the major vegetables consumed in Hong Kong and her neighbouring regions. It is of great value if the microspore culture system for Brassica parachinensis could be established. Accordingly, in this study factors including bud length, individual plant, culture induction temperature, sucrose concentration and pH value have
