Pharmacokinetics of intravenous and oral isosorbide-5-mononitrate

European Journal of Clinical Pharmacology, 1981
The pharmacokinetics of isosorbide-5-mononitrate (IS-5-MN) has been studied in two groups of healthy volunteers after oral (n = 20) and intravenous (n = 11) administration of 20 mg, which had previously been proved to be as effective as 20 mg sustained-release isosorbide dinitrate (ISDN).

Isosorbide 5-Mononitrate

Drugs, 1999
Imdur (IMD) is a sustained-release isosorbide 5-mononitrate preparation for the treatment of chronic stable angina pectoris. Controlled medication release is achieved using the Durules principle of insoluble matrix embedding. Data from randomised double-blind trials show that IMD 60 mg once daily (the most widely studied dosage) has significant ...

Isosorbide 5-Mononitrate Pharmacokinetics

Cardiology, 2008
Isosorbide 5-mononitrate differs from other clinically used organic nitrate vasodilators because of its almost complete oral absorption, the low intersubject variability in its plasma concentrations and pharmacokinetic parameters, its relatively long half-life and its lack of active metabolites.

Gas Chromatography-Mass Spectrometry Determination of Isosorbide 5-Mononitrate and Related Impurities in Raw Materials and Dosage Formulations

open access: yesJournal of Pharmaceutical and Biomedical Analysis, 1997
A straightforward quantitative method for gas chromatography-mass spectrometry determination of isosorbide 5-mononitrate (ISSMN) and its related impurities such as isosorbide (IS), isosorbide diacetate (ISDA) and isosorbide 2-acetate-5-nitrate (IS2A5N ...

Pharmacokinetic aspects of isosorbide-5-mononitrate in dogs.

The Journal of Pharmacology and Experimental Therapeutics, 1984
The aim of this study was to determine pharmacokinetic data of isosorbide-5-mononitrate (IS-5-MN) in dogs and to correlate them with the hemodynamic effects of this drug. Beagle dogs (n = 7) were given 3 mg/kg of IS-5-MN, the main metabolite of isosorbide-dinitrate, by i.v. injection and oral administration.

Inhibition of Angiogenesis, Tumour Growth and Metastasis by the NO-Releasing Vasodilators, Isosorbide Mononitrate and Dinitrate

open access: yesBritish Journal of Pharmacology, 1995
1. The effect of the nitric oxide (NO)-producing nitrovasodilators isosorbide mononitrate (ISMN) and isosorbide dinitrate (ISDN) were assessed on (a) the in vivo model of angiogenesis of the chick chorioallantoic membrane (CAM) and (b) on the growth and ...

New preparative routes to isosorbide 5-mononitrate

Journal of the Chemical Society, Perkin Transactions 1, 2000
Three new methods for the preparation of the vasodilator isosorbide 5-mononitrate are described. In the first method enzymatic regioselective acetylation of isosorbide gave isosorbide 2-butyrate. Nitration and deprotection of this material afforded isomerically pure isosorbide 5-mononitrate in high overall yield.

Isosorbide Mononitrate in Heart Failure With Preserved Ejection Fraction [PDF]

open access: yesNew England Journal of Medicine, 2015
BACKGROUND: Nitrates are commonly prescribed to enhance activity tolerance in patients with heart failure and a preserved ejection fraction. We compared the effect of isosorbide mononitrate or placebo on daily activity in such patients.

Treatment of Angina Pectoris with Isosorbide-5-Mononitrate

Cardiology, 2008
In this multicenter study 191 practitioners treated 532 patients with coronary artery disease and chronic stable angina pectoris by isosorbide-5-mononitrate (IS-5-MN), 20 mg 2–3 times daily. At the end of this period the frequency of anginal attacks was reduced by 83% and nitroglycerm consumption by 85.7% (p < 0.0001).

Eccentric dosing with isosorbide-5-mononitrate in angina pectoris

The American Journal of Cardiology, 1993
In a placebo-controlled, parallel, double-blind study, 214 patients with stable angina were studied. Doses of 5, 10 and 20 mg of immediate-release isosorbide-5-mononitrate (IS-5-MN) or placebo were administered at 0800 and 1500 hours for a period of 3 weeks.
