ITS-RFLP analysis, an efficient tool for differentiation of Bursaphelenchus species

Nematology, 2009
Abstract Restriction analysis of amplified ribosomal ITS sequences has provided species-specific fragment patterns for nematodes of several genera, including Bursaphelenchus. We used restriction enzymes RsaI, HaeIII, MspI, HinfI and AluI to produce ITS-RFLP reference profiles of 44 Bursaphelenchus species, including ...

Grouping of streptomycetes using 16S-ITS RFLP fingerprinting

Research in Microbiology, 2005
A total of 463 Streptomyces and Kitasatospora type strains were screened using 16S-ITS RFLP fingerprinting (combined restriction digest using enzymes BstUI and HaeIII). In total, 59 clusters could be delineated, each comprising multiple strains with nearly identical patterns. Good correlation was found in general with phylogeny, as revealed by 16S rDNA

PCR ITS-RFLP: A useful method for identifying filamentous fungi isolates on grapes

Food Microbiology, 2011
Restriction digestion analysis of the ITS products was tested as an easy method to identify isolates of filamentous fungi on grapes. Endonucleases SduI, HinfI, MseI, HaeIII were used. Endonucleases BfmI, Cfr9I, Hpy188I, MaeII or PspGI were used as necessary to complete discrimination.

The occurrence and rapid discrimination of Fomes fomentarius genotypes by ITS-RFLP analysis

Fungal Biology, 2012
Sequence comparison of available Fomes fomentarius (L.) J. Kickx f. internal transcribed spacer (ITS) of ribosomal DNA sequences demonstrated genetic non-homogeneity of the species. Multiple sequence alignment indicated the presence of two genotypes with overall similarity of about 97% and a strong statistics support.

ITS-RFLP fingerprinting and molecular marker for detection of Fusarium oxysporum f.sp. ciceris

Folia Microbiologica, 2010
Genetic diversity of 11 representative isolates of Fusarium oxysporum f.sp. ciceris causing chickpea wilt was determined through internal transcribed spacer (ITS) region of the ribosomal DNA-restriction fragment length polymorphism (ITS-RFLP). ITS1+5.8s+ITS2 regions of the isolates were amplified with a set of primers ITS1 and ITS4 and amplified ...

PCR ITS-RFLP for Penicillium Species and Other Genera

2016
Among numerous molecular methodologies developed for highly specific identification of filamentous fungi isolates, here we describe restriction digestion analysis of the ITS products as an easy method to identify isolates of filamentous fungi. This technique is a rapid and reliable method appropriate for routine identification of filamentous fungi ...

ITS-RFLP and ITS sequence analysis of a foliar endophytic Phyllosticta from different tropical trees

Mycological Research, 2003
Different isolates of a foliar endophytic species of Phyllosticta were isolated from different tropical tree species in India to examine genetic variation among the isolates. Internal transcribed spacer-restriction fragment length polymorphism (ITS-RFLP) analysis did not detect any variation among the isolates, suggesting that they all belong to the ...

Genetic relationships of pathogenic Typhula species assessed by RAPD, ITS-RFLP and ITS sequencing

Mycological Research, 2000
Several species of Typhula cause diseases of cereals and grasses at low temperatures. Taxonomic separation of T. phacorrhiza, T. incarnata and T. ishikariensis was investigated using RAPD, ITS-RFLP and nucleotide sequencing of the ITS region. Using 10 RAPD primers on genomic DNA or eight restriction enzymes on PCR-amplified ITS regions, banding ...
